Pancreatic tumor tissue was fixed in 4% PFA O/N i 4degC and embedded in paraffin. 5um sections were prepared and deparaffinized and rehydratated in Xylene and Ethanol (100% to 75%) twice each 5min and placed in ddH2O for 5min. Next, antigen unmasking was performed in citric acid based buffer (VectroLab #H3300) boil for 12min in microwave oven 600W. Let it cool down for 30min. Blocking in serum and avidin/biotin solutions was made. In many pancreatic cancers b-catenin activation and Wnt1 presence was noted.
