Anti-Versican 抗体 (ab19345)

Upon further investigation on how to use this antibody in IHC, I would like to recommend the following publications for your reference. We do not routinely test the antibody in this application, but have received customer reports that it works well.

I know you have had some questions about the immunogen and specifically what form of the protein is being recognzied. We are not experts in this field, but the originator was able to provide the following details:

This product is against the neoepitope of versican generated by cleavage of the versican core protein by ATAMTS 1/4. "The proteases ADAMTS-1/4 have been shown to cleave versican V0 and V1 in the GAG{beta} domain in vitro and in human aortic intima in vivo. This cleavage releases a 70-kDa N-terminal fragment from V1, thus severing the matrix cross-linking properties of the N- and C-terminal globular domains."

I have contacted the originator of this antibody and unfortunately I do not have any additional information regarding its use as a total measure of versican in the cell, as it will only recognize the cleaved version. It may be useful to search the primary literature to learn more about how the detection of the cleaved protein translates into the analysis of total protein expression.

Leider habe ich eine schlechte Nachricht für Sie: Aggrecan hat insgesamt eine Übereinstimmung von 19%, es gibt eine Region zwischen AS 800-850 des menschlichen Proteins, welches eine hohe Übereinstimmung mit dem Ziegen Protein hat, nur haben wir leider keinen passenden Antikörper.

Versican und SOX5 weisen leider gar keine Übereinstimmung mit dem Ziegenprotein auf.

Da alle unsere Aggrecan,Versican und SOX5Antikörper gegen das humane Protein entwickelt worden sind, kannich Ihnen leider keinen Vorschlag machen.

Please find attached the link which actually explains the reason of band at ˜70-80 kDa. This is the cleavage product of versican. Please refer to fig 1.

The CGG residues are part of the immunogen but not of the protein itself and the immunogen sequence matches to the isoform V1 of human versican protein,residues436-441. The anti-Versican antibody ab19345does not detect the full-length (266 kDa) protein, but only the 70 kDa N-terminal ADAMTS cleavage product of versican V1. The proteases ADAMTS-1/4 cleave versicanV1 in the GAG domain in vitro and in human aortic intima in vivo. This cleavage releases a 70 kDa N-terminal fragment from V1, thus severing the matrix cross-linking properties of the N- and C-terminal globular domains. I hope this information is helpful to you.
