      Immunohistochemical analysis of HIV-infected syphilitic skin tissue, staining Treponema pallidum with ab20923.

      Antigen retrieval was performed before blocking with normal horse serum and 2% BSA. Samples were incubated with primary antibody overnight at 4°C. A biotinylated horse anti-rabbit IgG was used as the secondary antibody. Staining was detected using AEC.

    Inquiry: We are interested in purchasing this antibody for IHC on formalin-fixed paraffin-embedded tissue, but before it is approved by our head of department I need to get an idea of what the working dilution might be (so we can estimate costs). We use the Roche Ventana ultraView detection kit and Benchmark XT and Ultra staining platforms. Do you have any experience in using this antibody with these reagents? If not, can you give me an idea of the dilution used with a standard SAB technique and I will use this as a starting point. Thanks for your help.

    We have unfortunate not used the anti-Treponema pallidum antibody (ab20923) with the reagents you have described. I can say we have used the antibody in a 1/100 dilution in staining of paraffin embedded tissue sections. However, this is likely to require further optimisation when used in your particular tissues and protocol to obtain the best results.

    I would just like to point out that the reagents we provide are all intended for research and development purposes only. They are not suitable for diagnostic or clinical purposes.

    We have a customer who has recently purchased the above antibody and they have a questions with regards to the positive control;



    I recently purchased the abcam anti-T pallidum antibody cat#AB20923. I am planning to work up this Ab for IHC-P (IPX)



    I have so far been unsuccessful in finding positive paraffin tissue samples in our files. Do you know if abcam or any other company sells positive paraffin section controls for syphilis?

    Unfortunately, we do not sell a positive control slide either for this antibody.

    I cannot guarantee that this is a reliable source for the slides, but I found a company via the internetwhich can actually slides which are suitable for immunohistostaining and not only for Silver Staining:
