    Receptor with high affinity for TNFSF2/TNF-alpha and approximately 5-fold lower affinity for homotrimeric TNFSF1/lymphotoxin-alpha. The TRAF1/TRAF2 complex recruits the apoptotic suppressors BIRC2 and BIRC3 to TNFRSF1B/TNFR2. This receptor mediates most of the metabolic effects of TNF-alpha. Isoform 2 blocks TNF-alpha-induced apoptosis, which suggests that it regulates TNF-alpha function by antagonizing its biological activity.

      Intracellular Flow Cytometry analysis of Jurkat (Human T cell leukemia T lymphocyte) cells labeling TNF Receptor II with Purified ab109322 at 1/20 dilution (10 µg/mL) (Red). Cells were fixed with 4% Paraformaldehyde and permeabilised with 90% Methanol. A Goat anti rabbit IgG (Alexa Fluor® 488, ab150077) secondary antibody was used at 1/2000. Isotype control - Rabbit monoclonal IgG (Black). Unlabeled control - Cell without incubation with primary antibody and secondary antibody (Blue).

      Immunocytochemistry analysis of Jurkat (Human T cell leukemia T lymphocyte) cells labeling TNF Receptor II with Purified ab109322 at 1/50 dilution (2.66 µg/mL). Cells were fixed in 4% Paraformaldehyde and permeabilized with 0.1% tritonX-100. Cells were counterstained with Ab195889 Anti-alpha Tubulin antibody [DM1A] - Microtubule Marker (Alexa Fluor® 594) 1/200 dilution (2.5 µg/mL). Goat anti rabbit IgG (Alexa Fluor® 488, ab150077) was used as the secondary antibody at 1/1000 dilution (2 µg/mL). DAPI (blue) was used as nuclear counterstain. PBS instead of the primary antibody was used as the secondary antibody only control.

Perform heat mediated antigen retrieval using ab93684 (Tris/EDTA buffer, pH 9.0). ImmunoHistoProbe one step HRP Polymer (ready to use) was used as the secondary antibody. Negative control: PBS instead of the primary antibody. Hematoxylin was used as a counterstain.

    Please let me know whether any of the cell lines you used is transfected with NTFRII (thus overexpressing it). Also, let us know how many micrograms of extract you loaded on each lane of the blot you sent us.

    I've heard back from the lab, and they load 10 ug of protein per lane. None of the cell lines are transfected, and they use the general Epitomics Western blot protocol-

    I've asked the lab whether the cells are transfected, and how many micrograms of lysate were loaded.

    I'd be happy to send a positive control lysate, however I can't send all of the lysates shown in the image. We have the RAW 264.7 lysate ab7187 or NIH/3T3 lysate ab7179-

    I've received the attached image from the lab, which shows Western blot results using C6, Raw264.7, PC12, and NIH/3T3 cell lysates.

    The band probably aligns most closely with the ˜ 60 kDa band that you described, but please let me know if testing with the alternative antibody ab119998 would be helpful, and I'll send this out to you.
