If you continue to have problems with the antibody please also let me know. As discussed in my previous email, there are a few protocol changes that I think would be worthwhile trying in order to see if the antibody performs any better with your samples.

From having reviewed the protocol details provided there are q few thinks I would suggest would be worthwhile to try to obtain the best staining with the antibody.

1. I would suggest increasing the concentration of antibody used, trying 1/100 to start with to see if any improvement is seen.

2. It may be worthwhile to try and optimise the antigen retrieval used. Applying sodium citrate buffer for 30 minutes bay be a little too long. I would suggest treating the tissue with boiling citrate buffer (using microwave or pressure cooker) for 5, 10 and 20 minutes to see which gives the best results.

I do however understand that you have spent some time in optimising the conditions and you have used the antibody previously. I would therefore be happy to provide you with a replacement vial of ab14692 (of a different lot number), or an alternative antibody altogether such as rabbit monoclonal https://www.abcam.com/Synaptophysin-antibody-EP1098Y-ab52636.html.

If you would like for me to arrangeeither of these options, could you pleaseconfirm the delivery address andbilling address (as we only have that of ATOM which placed the order originally).

1. Which type of tissue samples are being stained? Human? Mouse? From which organ?

2. Has the secondary antibody been used successfully before?
