    S.aureus binds IgG molecules by their Fc region. In serum, the bacteria will bind IgG molecules in the wrong orientation on their surface, which hinders opsonization and phagocytosis. Mutants of S. aureus lacking protein A are more efficiently phagocytosed in vitro, and mutants in infection models have diminished virulence. Due to its affinity for the Fc region of many mammalian immunoglobulins, protein A is considered a universal reagent in biochemistry and immunology.

      ab20920 staining an abscess in rat calf tissue prepared as a 20 µm cryoslice.  Tissue was fixed in paraformaldehyde, permeabilised with 0,5% Trition X100 in PBS, and blocked with 10% serum plus 1% BSA for 1 hour at room temperature.  The primary antibody was diluted 1/500 with PBS plus 3-5% Serum, 0,5% Trition X100 and incubated for 24 hours at 20°C.  An Alexa Fluor® 568 goat anti-rabbit was used as the secondary

      ab20920 staining Staphylococcus aureus in human keratinocytes by Immunocytochemistry/ Immunofluorescence.
      Cells were fixed in methanol, permeabilized using 0.1% saponin/ PBS, blocked with 4% BSA for 30 minutes at 25°C and then incubated with ab20920 at 10µg/ml for 14 hours at 4°C. The secondary used was an Alexa-Fluor 488 conjugated goat anti-rabbit polyclonal used at a 1/100 dilution.

    Answer

    The cells used for the immunogen started out as whole cells that were disrupted by sonication and freeze/thaw cycles. Therefore the immunogen is a mixture of intact Staph aureus organisms and soluble proteins from disrupted cells. This is no record of UV-inactivation.

    There is absolutely no problem, the antibody will still work properly in my opinion.
    Indeed, we have tested different antibodies, and even after a week at 37 degrees Celcius, the antibodies did not seem to loose activity. I am therefore confident that the antibody ab20920 you received, will still work perfectly fine.
    If against expectations the antibody would not work, do please contact us again and we will send a replacement vial.

    Please find below the information I managed to collect by contacting the sources of the Anti-Staphylococcus aureusand the Anti-Streptococcus agalactiaeantibodies we have in our catalog :

    Anti-Staphylococcus aureus:
    https://www.abcam.com/ab23568 shows no reactivity to any streptococcus sp., mycoplasma sp. or corynebacteria sp.

    Unfortunately, ab35192 has not been specifically tested against streptococcus so we are unable to guarantee it won’t react to S. agalactiae.

    ab68950, ab68952, ab68954: These Staphylococcus aureus polyclonal antibodies are not absorbed for related microorganisms and so therefore could cross react with related bacteria such as Staph. Epidermidis.

    MLS : Staphylococcus aureus: We do not have an antibody that is specific to Staph. aureus but does not have crossreactivity to Staph. epidermidis.

    Anti-Streptococcus agalactiae:
    https://www.abcam.com/ab21126 has weak crossreactivity with Strep. dysgalactiae and does not cross-react with Strep. uberis, Strep. bovis, Strep. faecalis, Staph. aureus, Mycoplasma bovis, Mycoplasma californicum, Mycoplasma bovigenitalium, E. coli or Corynebacterium bovis.

    Thank you for contacting us. Indeed, neither ab68954 nor ab20920 have been tested for immunoprecipitation (IP) yet, but they could theoretically be used in this application. If it is just between the two, I would recommend ab20920 because it is an unconjugated antibody. The biotin-labeled antibody ab68954 might not bind to the Protein A or G agarose as well as the unconjugated antibody. Also, ab20920 seems to detect its antigen(s) in a rather native state (as it works with frozen tissue sections) which might be more suitable for IP. However, these antibodies are polyclonal antibodies against various soluble and structural antigens of the whole Staphylococcus aureus. By using the antibodies in IP, a variety of proteins would be pulled down. If this is not appropriate for your experimental setting, I would be pleased to assist you in finding a suitable antibody (for example against a certain target protein). If you decide to go ahead and purchase one of these products though, you might be eligible for our testing discount program.
