Sperm cells were fixed with 2% EM grade formaldehyde and storage in 4oC (refrigerator). Approximately 500,000 sperm in 100 µL of PBS were incubated at room temperature with 5 µL of antibody anti-SPINK6 at a concentration of 0.1 µg/µL conjugated to PE/CY5® (ab102893). Antibody binding was accessed every hour starting at the 1st hour of incubation until the 4th hour, a final assessment of antibody binding was conducted at the 24th hour of the incubation. Several samples were tested, and no binding was achieved.
