      Lane 1 : Rat enterocytes
      Lane 2 : Rat heart tissue
      Lane 3 : Human skeletal muscle tissue

      Predicted band size: 73 kDa



      Tissue extracts were made in Solobuffer with protease inhibitors. The samples were solubilized in SDS-PAGE sample buffer and reduced with 2.5% BME.
      Total 40-50ug protein was applied per lane on 10% SDS PAGE gels.
      The proteins were transferred to nitrocellulose F and blocked with 5% milk and blotted with 1:500 diluted ab14686. 

      The image was obtained with high sensitivity ECL assay and Image was captured with a CCD cooled camera for 30 sec.

    Answer

    Thank you for your enquiry. I am afraid that there is no further information regarding the positive control that what is already stated on the datasheet. "The positive control for Sglt1 was purified rat jujenal brush border membranes solobilized in Solo buffer at approximately 1.5 mg per ml. A 65 ul sample was applied on 8.5% gel, the approximate MWt of rat Sglt varies between MW of 70-74kda depending upon the concentration of acrylamide in the gel." However, for your information the actual positive control used was product ab99447 SGLT1 protein. I hope this information will still be helpful.

    Dear Sir/ Madam, Sorry to disturb. Our customer raises an inquiry between ab14685 and ab14686 (both Anti-SGLT1 antibody). She would like to conduct WB with rat sample, and these two products meet her need. The predicted protein size is around 73kDa in the human as well as the rat species. But she is confused about the detected molecular weight in the datasheet of ab14685: “Detects a band of approximately 55-62 kDa”. She therefore would like to confirm the actual molecular weight in Rat sample when using these 2 antibodies. Would you please help her deal with this issue? Thank you very much. Beat regards,

    Thank you for your inquiry. I can confirm that rat SGLT1 has a predicted  molecular weight of ca. 73kDa. Molecular weights on gel are always only a rough estimate since the gel and buffer conditions can influence the resulting molecular weight of a protein dramatically. It is know in this case that also the percentage of the gel has an influence on the molecular weight of SGLT1. I hope i could reassure your customer.     Hello, I´m trying to perform a Western Blot for Human SGLT1(using anti-SGLT1 ab14686), but I have problems with the blotting. I think there´s a problem with the cellular lysis. I already tried to use different lysis buffer (e.g. RIPA, Rotiload) and different temperatures, but either the samples remain in the pockets of the gel or I detect a wrong size (aroud 40 kDa). Can you please write me the protocol you used or give me any suggestions? Thank you very much. Kind Regards,

    I am sorry to hear that you have been experiencing problems using this product in the application that you wish. It is known that more than 10 types of glucose transport carrier proteins designated as Glut 1-13 facilitate glucose transport across the cell membrane. Molecular cloning of glucose transporters have identified a family of closely related genes that encodes at least 7 proteins exhibiting high degree of amino acid homology (45%-65%) all in the molecular weight range of 40-60 kDa. Please could you provide some further details of the protocol used and complete the following form (attached as a word document).
