Antigen Retrieval: Bring slides to a boil in 10 mM sodium citrate buffer (pH 6.0), then maintain at a sub-boiling temperature for 10 minutes. Cool slides on bench-top for 30 minutes. Block each section with 100-400 ul blocking solution (1% BSA in PBS) for 1 hour at room temperature. Incubate with ab15093 overnight at 4C. |

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-SCP3 antibody (ab15093)Image is courtesy of an anonymous AbReviewab15093 staining SCP3 in Human TEFNA testis tissue sections by Immunohistochemistry (IHC-P - paraformaldehyde-fixed, paraffin-embedded sections). Tissue was fixed with paraformaldehyde and blocked with 5% serum for 30 minutes at 25°C; antigen retrieval was by heat mediation in a citrate buffer. Samples were incubated with primary antibody (1/200 in PBS + 5% FCS + 0.1% Triton X-100) for 1 hour at 25°C. Ab6721 (1/1000) was used as the secondary antibody.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-SCP3 antibody (ab15093)Ab15093 staining human normal testis tissue. Staining is localized to nuclear compartment.

Sections were stained using an automated system DAKO Autostainer Plus , at room temperature. Sections were rehydrated and antigen retrieved with the Dako 3-in-1 AR buffer EDTA pH 9.0 in a DAKO PT Link. Slides were peroxidase blocked in 3% H2O2 in methanol for 10 minutes. They were then blocked with Dako Protein block for 10 minutes (containing casein 0.25% in PBS), then incubated with primary antibody for 20 minutes, and detected with Dako Envision Flex amplification kit for 30 minutes. Colorimetric detection was completed with diaminobenzidine for 5 minutes. Slides were counterstained with Haematoxylin and coverslipped under DePeX. Please note that for manual staining we recommend to optimize the primary antibody concentration and incubation time (overnight incubation), and amplification may be required. -
