    Abcam's Sarcosine Assay Kit provides an accurate, convenient measure of sarcosine in variety biological samples. In the assay, sarcosine is specifically oxidized to generate a product that converts a colorless probe to a product with intense red color (λmax = 570 nm) and which is also highly fluorescent (Ex/Em = 538/587 nm). Sarcosine is therefore easily detected by either colorimetric or fluorometric methods with detection range 1-10000 µM.

    Sarcosine (N-methylglycine) is an intermediate and byproduct in the synthesis and degradation of glycine. Sarcosine is metabolized to glycine by sarcosine dehydrogenase, while glycine-N-methyl transferase generates sarcosine from glycine. Sarcosine is a natural amino acid found in muscles and other body tissues and plays important role as an intermediate in the metabolism of choline, methionine, glycine, glutathione, creatine, purine and serine.

    Thank you for your enquiry. Please find below the answers to your questions. 1. Biospecimens like body fluids (plasma/ serum), tissues, and cell culture samples can be used. However, urine samples will give interference with the assay. For cell and tissue sample you may follow the procedure mentioned below: For cell samples: Start with 2X106 cells, suspend the cell pellet 500ul (~4 volumes) of the assay on ice, pass it through the Dounce homogenizer (10-50 times) on ice, till efficient lysis is confirmed, by viewing the cells under the microscope. Spin down the sample and then use the supernatant for your subsequent assays. For tissue samples: Start with 10mg of the tissue, add 500-1000 ul (~4-6 volumes) of the assay buffer, pass it through the Dounce homogenizer (10-50 times) on ice, till efficient lysis is confirmed, by viewing the cells under the microscope. Spin down the sample and then use this supernatant for your subsequent assays. 2. Ideally it is better if you use fresh sample for your assay. Samples that have been appropriately stored at -20 C or -80 C should also be stable for about two months for this assay. 3.
