      we used BSA as well, this did not make a difference. The
      unspecific bands are more pronounced in the total cell
      lysates than in the IP samples. Further washing steps did
      not change the amount of unspecific bands either. We use
      human epidermal cells as well as Hek293 and HeLa cells. The
      bands appear in all cell lines. The other vials were all
      working properly.

      Having reviewed this case, I would like to offer some suggestions to help optimize the results from ab101349. I would also appreciate if you can confirm some further details:

      - Use BSA instead of milk, this can sometimes give more satisfactory results and alter background signal.
      - Incubate the primary antibody and membrane at room temperature for 6-8 hours. This can increase the level of specific antibody binding.
      - Do you see different background in the IP'd and non-IP'd samples?
      - What species and cell lines have you tested?
