    Respiratory syncytial virus (RSV) is a major cause of respiratory illness in young children. RSV infection produces a variety of signs and symptoms involving different areas of the respiratory tract, from the nose to the lungs. RSV is a negative sense, enveloped RNA virus. The virion is variable in shape and size with average diameter of between 120 and 300 nm. The 63 kD RSV fusion protein of the RSS 2 strain (subtype A) directs fusion of viral and cellular membranes, results in viral penetration, and can direct fusion of infected cells with adjoining cells, resulting in the formation of syncytia or multi nucleated giant cells.

    Thank you for your enquiry. I am sorry to hear that Abcams Respiratory Syncytial Virus antibody (ab12253) is not proving a success. Please can I ask some follow up questions. What kind of result did the RSV-infected Hep-2 cells that you are using as a positve control give. Was it a strong signal? Is it a cell lysate that you are using that has previously been shown to give a result, or cells harvested following transfection? I think it would be of benefit to perform this WB one more time using a cell lysate that you are sure contains the target protein. It would help if you could perform this in parallel with the goat anti-RSV polyclonal antibody previously shown to work by WB. Please can you leave a space between the MR markers and the postive control. So in parallel perform WB on 1. MR markers, 2. BLANK, 3. POSITIVE LYSATE (20ug), 4. POSITIVE LYSATE (10ug), 5. NEGATIVE LYSATE (20ug). Please do not hesitate to get back in contact with me should you require further assistance.

    Thank you for contacting us for technical support, I'm sorry to hear you are experiencing a problem with ab12253. The following suggestions will help you increase your signal: -heat the samples at 95-100C for 5min to make sure the protein is all denatured and can be detected easily by the antibody in this form -block 1hr only in 5% milk in TBST -incubate the primary antibody overnight at 4C (in TBST) -use a more sensitive detection system, such as ECl+ or pierce super west pico signal kits. -please make sure the secondary antibody works with other primary antibodies and is not old or damaged. Please let me know if you still experience problems with those changes and I will arrange a refund or replacement vial if you purchased the antibody in the last 90days.
