Thank you once again for your enquiry. I am pleased to forward you some futher procedure details for testing of ab in cell culture. I hope this will be helpful to you. 1. Resuspend 3T3 cells (passage#38) in regular culture media with 10% calf serum. Transfer 100ul culture/well to assay plate (5,000 cells/well). Incubate overnight. 2. Complete media change. Replace with 100ul media containing 0.5% calf serum. Incubate overnight. 3. Complete media change. Replace with media containing 0.5% calf serum. 4. Serial dilute h-TGF-alpha in media with 0.5% calf serum. 5. Transfer 100ul diluted cytokine cells to assay plate. Volume of each well is 200ul, containing 0.5% calf serum and h-TGF-alpha. Incubate 45 hours. 6. Add 20ul of Substrate Cell Titer 96 Aqueous One Solution Reagent to each well. 7. Incubate 37oC and read at OD 490nm.
