    Enzyme that cleaves 2',5'-phosphodiester bond linking adenosines of the 5'-triphosphorylated oligoadenylates, triphosphorylated oligoadenylates referred as 2-5A modulates the 2-5A system. This enzyme degraded triphosphorylated 2-5A to produce AMP and ATP. Also cleaves 3',5'-phosphodiester bond of oligoadenylates. Play a role as a negative regulator of the The 2-5A system that is one of the major pathways for antiviral and antitumor functions induced by interferons (IFNs). Suppression of this enzyme induces reduction of viral replication in Hela cells, thus counteracting the antiviral pathway probably by inhibiting the 2-5A system.

Thank you for your patience and for your feedback. I am very sorry to hear that the new protein does also not show enough activity for being used in an activity assay. In regards to use the whole cell extract, I am not very optimistic to be honest. I am not sure whether the lab is able to provide it, and even if so, it would be complicated and take time. More importantly however, I am not very hopeful that the lysate would have activity if the purified protein does not show activity.  and I do not have much hope that the transfected cells would give a better result. Indeed, in the cell extract there are many additional factors which could inhibit the activity of the protein. I am sorry for not being able to be more optimistic on this. I would actually suggest to search for a PDE12 protein produced in a different system. I have asked my colleagues to check with their collaborators whether there might be another PDE12 protein available - maybe even a PDE12 protein tested for functional activity. I will let you know as soon as I have received the answer. Meanwhile, can you please also submit the Abreviews for the two proteins? We would greatly appreciate if you could put there also a summary of the test procedure in the additional notes.  Thank you very much! Please let me know your thoughts and if you have any questions. Review text: Dilutions of PDE12 (1 to 10ml) were incubated for 2h with 20µl of PDE12 assay substrate, an analogue of 2-5A dual labelled with Fam and Dabcyl. Active PDE12 is expected to cleave 2-5A molecules releasing the quencher from the Fluorophore, allowing it to fluoresce which is detected by the Flo-Star Optima (Ex 485nm and Em 520nm). Delta fluorescence (T120min-Tzero min) is used to calculate PDE12 % activity using cell lysate as positive control for PDE12.

Unfortunately there was no detectable fluorescence (Tzero and T120 were almost the same) observed after incubating PDE12 (ab107695) for 2h suggesting the protein under the storage conditions provided was not active.
