Specific Activity: 0.096 pmol/min/μg.Assay Conditions: 10 µl reaction mix containing assay buffer with 50 mM HEPES (pH 7.4), 1 mM TCEP, 100 µM alpha-ketoglutarate, 25 µM iron, 0.1 mM ascorbic acid, 0.02% Tween-20, 0.5 µM biotinylated peptide substrate, and ab271785 (10 – 50ng) added to the wells. Add antibody against di-methylated K27 peptide. Incubate for 30 min, then Streptavidin-conjugated secondary antibody followed by Alpha Screening detection. -

Histone demethylase that specifically demethylates 'Lys-27' of histone H3, thereby playing a central role in histone code. Demethylates trimethylated and dimethylated but not monomethylated H3 'Lys-27'. Plays a central role in regulation of posterior development, by regulating HOX gene expression. Demethylation of 'Lys-27' of histone H3 is concomitent with methylation of 'Lys-4' of histone H3, and regulates the recruitment of the PRC1 complex and monoubiquitination of histone H2A. -
