      HIV1 performs highly complex orchestrated tasks during the assembly, budding, maturation and infection stages of the viral replication cycle. During viral assembly, the proteins form membrane associations and self-associations that ultimately result in budding of an immature virion from the infected cell. Gag precursors also function during viral assembly to selectively bind and package two plus strands of genomic RNA. Capsid protein p24 probably forms the conical core of the virus that encapsulates the genomic RNA-nucleocapsid complex.

    The immune-biological activity of HIV p24 is derived from its linear AA sequences and has no secondary structure related activity. This protein has not been tested for functional activity.

    The product is in 8M urea for the stock solution. My concern was, when the antigen is diluted to pg/mL level with a buffer without urea, is this antigen still stable or remain in the conformation that can be detected by ELISA?

    You are correct, ab43037 is an antigen, not an antibody, and it is suitable to be coated on an ELISA plate from 100 ng to 10 ug. The dilution buffer that you use does not have to contain urea, as long as you are coating the wells within the range given above.

    Question

    If this antibody is diluted further so the Urea concentration is low enough to not interfere with an ELISA, would the protein still be stable? I am concerned about the high Urea concentration interfering with the ELISA, so I want to make sure I dilute it sufficiently.

    I have checked some of the HIV1 p24 proteins available in our catalogue with my lab colleagues. Regarding their suitability for ELISPOT, I am afraid it hasn’t been tested in our lab, but due to their suitability in ELISA, they should also work in ELISPOT.
    As an example, ab127888 is highly specific in ELISA, but ab73768 or ab43037 are also very likely to work in this assay.
    If you would be interested, you could participate in our Antibody evaluation program, by which you test an antibody in a new species/application (such as any of the referred antibodies in ELISPOT), and share the results with us (submitting an Abreview), and you beneficiate from a free antibody.

    Thank you for your reply. it is very helpful.   The research I will carry out is to apply the magnetic detection echnique into HIV detection through sandwich assay.   From the information you provide, HIV1 p24 protein (ab73768) is the antigen I need, right? Since it can be detected by all three antibodies  Anti-HIV1 p24 antibody [ND1] (ab8246) and  Anti-HIV1 p24 antibody [ND1] (FITC) (ab81433) and Anti-HIV1 p24 antibody [39/5.4A] (ab9071).   I wonder if the ab81433 is the same as ab8246 except the ab81433 has FITC fluorogen which could be detected by fluoresence absorption. Right?      Looking for your reply. Thank you!

    Thank you for your reply. Unfortunately, I would not recommend the protein ab73768. This is only a fragment of the HIV1 p24 that lacks large parts of the protein such as the N-terminal amino acids (1-76). If the antibodies you would like to test recognise an epitope within these missing parts (which we do not know as they have not been mapped), these antibodies would not be suitable for the proposed assay. Therefore, I would recommend to use the protein ab43037 which is the full-length HIV1 p24 and will be detected by the antibodies. Indeed, I can confirm that ab8246 and ab81433 are the same antibody clone, thus detecting the same epitope, and that ab81433 is conjugated to FITC which can be used in applications such as immunocytochemistry/immunofluorescence (ICC/IF). For your experiments it seems that you do not need the fluorochrome, so I would recommend ab8246. In summary, for the sELISA two antibodies that recognise different epitopes are required and the matching pair needs to be determined by the end user. To our knowledge, neither ab43037 nor ab8246/ab81433 nor ab9071 have yet been tested in sELISA.     Thank you for contacting us. The differences are as follows: ab43036 HTLV I p24 protein: This protein is the full length sequence of HTLV I p24, i.e. an in E. coli derived recombinant protein. The Human T-lymphotropic virus Type I (HTLV-1) is not identical with HIV. When Human immunodeficiency virus (HIV), the virus that causes AIDS, was characterized, it was originally named HTLV-III. Therefore, this protein seems not suitable for your experiments. ab43037: This protein is the recombinant full length protein HIV1 p24, expressed in E.coli. This protein is guaranteed to work in Western blot and ELISA. ab73028: This protein is the recombinant full length HIV1 p24 from HxB2 strain purified from Pichia pastoris. It contains a 6x histidine fusion partner which for example facilitates purification. ab73768: This protein is only a recombinant fragment (amino acids 77-436) of the HIV1 p24 protein and is additionally fused to beta-galactosidase. If you could provide some more information on the type of assay you would like to perform and let us know whether you are interested in HIV-1 or HIV-2, I would be happy to make a specific recommendation. In case the assay is similar to a sandwich ELISA (sELISA), I can let you know that we have tested neither the proteins nor any antibodies for their suitability in sELISA, as this has to be tested for each pair individually.
