Good afternoon,

we would like to ask for help regarding the next secondary antibodies, guiding me through the choice to use some antibodies for Immunofluorescence in combined forms.

The target cells are VERO, infected with a porcine virus.

Some of the primary antibodies have then to be tagged with a goat anti-mouse Cy3 or a donkey anti-mouse FITC.

To this primary anti-mouse marking we perform another signaling mark with a rabbit anti-pig (ab6775 - already in our lab) or one of the following items:

rabbit a-pig ab6773 (FITC)
goat a-pig ab6911 (FITC), Which should result better? Should we be careful about the Fc portion in such combinations?

To add up to this confusing situation we also have a need for a anti-rabbit Dylight 594. Should we prefer a donkey anti-rabbit (ab 96921 or 96893) or,
a goat anti-rabbit (ab 96885, 96897 or 98473)?

Thank you very much for your inquiry and your interest in our products.

If performing a double or triple staining, it is very important to make all the controls that the secondary antibodies do not crossreact with the immunoglobulins from the other species. For example for the anti pig antibody, it would be important that the anti pig antibody does not recognise the immunoglobulins from mouse or rabbit, not to have a false collocatisation. Unfortunately, non of our anti pig antibodies have been tested for this. However as you have used the ab6775 already successfully in your laboratory if I understood correctly, I can recommend to use the ab6773 as it is from the same original pool. Please do however perform the single color controls to verify the crossreactivity of the ab6773 with rabbit or mouse immunoglobulins.

In regards to the anti rabbit antibody, I can recommend to use the ab98500, a donkey anti rabbit IgG which has been preabsorbed against mouse, rat and human immunoglobulins, sothere is nocrossreaction against those immunoglobulins. We do not have an anti rabbit DyLight 594 which has been crossabsorbed also against the pig immunoglobulins. If you are concerned or if you have troubles with background staining on the VERO cells, I can recommend to use the same antibody , however with no Fc region, i.e. the F(ab')2 portion. This would be the ab98490.

The species in which the secondary antibody is generated is not very important. It is only important if you get background staining from the secondary antibody alone, in which case you can then block with serum of that species. However this is not an issue in general, especially not in cell culture.
