I have been waiting to obtain more details on the immunogen for this antibody from the laboratory. I can confirm that the immunogen sequence was from amino acids 367-388 of EP3. This corresponds to cytoplasmic C-terminal end of the protein. I can therefore recommend adding a permeabilisation step to your staining procedure. As it is a membrane protein, I can suggest using a mild detergent, such is digitonin or saponin at 1% (Triton will be too harsh). I can also recommend that as you are detecting a recombinant protein, which are sometimes shorter than full length protein, it is important to ensure this includes the immunogen sequence. I can suggest this can be checked with the immunogen details above.
