    The proteasome is widely recognised as the central enzyme of non lysosomal protein degradation. It is responsible for intracellular protein turnover and it is also critically involved in many regulatory processes and, in higher eukaryotes, in antigen processing. The 26S proteasome is the key enzyme of the ubiquitin/ATP dependent pathway of protein degradation. The catalytic core of this unusually large (2000 kDa, 450Å in length) complex is formed by the 20S proteasome, a barrel shaped structure shown by electron microscopy to comprise of four rings each containing seven subunits. Based on sequence similarity, all fourteen 20S proteasomal subunit sequences may be classified into two groups, alpha and beta, each group having distinct structural and functional roles. The alpha subunits comprise the outer rings and the beta subunits the inner rings of the 20S proteasome. Observations of the eukaryotic proteasome and analysis of subunit sequences indicate that each ring contains seven different subunits (alpha7 beta7 beta7 alpha7) with a member of each sub family represented in each particle. Each subunit is located in a unique position within the alpha or beta rings. 20S Proteasomes degrade only unfolded proteins in an energy independent manner, whereas 26S proteasomes degrade native and ubiquitinylated proteins in an ATP dependent manner. The native protein substrates are recognised by subunits, some with ATP binding sites, of the outer 19S caps of the 26S proteasome.

    ab22673 staining human Hela cells by ICC/IF. Cells were formaldehyde fixed, permeabilized in 0.1% Triton and blocked in 3% BSA for 15 minutes at room temperature. The primary antibody was diluted 1/2000 (3% BSA) and incubated with sample for 1 hour. An Alexa Fluor® 488 conjugated goat polyclonal to rabbit, diluted 1/1000 was used as secondary.

    Immunofluorescence analysis of HeLa cells, staining Proteasome 20S alpha + beta with ab22673.

    Cells were fixed with formaldehyde and permeabilized using 0.1% Triton X-100. Samples were incubated with primary antibody (1/1000) fr 1 hour at room temperature. An AlexaFluor®555-conjugated donkey anti-rabbit IgG was used as the secondary antibody.

I can change the antibody on your current order from ab22673 for ab109525 if you would like. As this is in stock, if you let me know you would like for me to do this, it can be with you by tomorrow.

The antibody has been used both in WB and IF and should therefore be suitable for your experiments. Please be aware that this antibody is raised against the Proteasome 20S alpha 2 and will detect this protein. It has been shown to work with mouse, rat and human samples.

Thanks for your kindly reply, sorry about the error context, after I contacted with this customer she correct the context and replied you question as follow:

1st antibody 4 degree overnight

Ab22673 (proteasome,α,and,β,subunits):,1:1000 in 3% BSA,

Tuj-1: 1:1000 in 3%BSA

2nd antibody 3hrs RT

Anti Rabbit AlexFluo488:1:5000 in 3%BSA

Anti chicken AlexFluo 543: 1:5000 in 3%BSA

Fixation:4% PFA in PBS for 12 mins RT

Permeabilization:0.3% triton-X100 in PSA for 12 mins RT

The proteasome staining is green channel and the other antibody is anti-taj-1(red), and we expect to see the signal in cortical neurons.

This customer indicated she also used the other primary antibody to conducted the same experiment, but the result seems well,

therefore, she think this antibody lost its function already, and she also wants to ask for your kindly help to create a credit note to her.

I also attached the image in this letter. Please check it and could you please help this customer to solve the problem?

The results show non-specific staining; therefore, she wants to ask for your help to modify her experiment step, could you please offer any suggestion to help her?

I also attached her data in this letter and her experiment step as follow:

Thank you for contacting us and forwarding the customer's protocol details.

I do have a few additional questions, so that I better understand the situation.

The secondary is listed as HRP conjugated, and the detection system is listed as ECL, however the image shows fluorescent deteciton.Is the proteasome stainingin the green or red channel? What other protein has been stained, and what kind of antibody was used?

I've looked through some literature about the expression of this protein, and the subunits can be found in the nucleus or cytoplasm. What kind of staining does the customer expect to see in these hippocampal and cortical neurons?

The customer's protocol looks good, but to reduce background I would recommend blocking the cells with 10% serum from the host of the secondary antibody (10% goat serum in this case) which often improves the results. We do fully guarantee this antibody to work in rat ICC, so if the customer would prefer, I'd be happy to send a replacement or issue a credit or refund.
