    Abcam's Genomic DNA Isolation Kit provides a simple and convenient procedure for rapid isolation of genomic DNA from mammalian cells and tissue samples with high yield and purity. The novel method requires less than 90 minutes to prepare highly pure genomic DNA. The extracted genomic DNA is free from protein and RNA, and suitable for a variety of applications such as PCR, DNA hybridization, enzyme manipulation, cloning, Southern blot, and array-based experiments.

Question

I have some questions about this genomic DNA isolation kit:
1. When resuspending the enzyme, there was a hazy solution that remained because not all the lyophilized material was resuspended. Is this normal? What is one supposed to do here?
2. My OD260/280 in to preps was 1.5 and 1.6. I require at least 1.7. Could the enzyme not being completely resuspended have an effect here?
3. After washing the pellet with 70% ethanol and drying for 5', adding TE did not resupend the entire pellet. It may have shrunk a little upon addition of the TE but it was not all suspended.

Answer

We always provide more material than necessary which sometimes results in a super saturated solution. This can lead to a hazy solution. Typically this does not compromise function in any way. You can warm the solution up to 37C or use mild sonication in a water bath for 1-2 mins to try to dissolve all the material. If there is still undissolved material I would recommend simply proceeding with it.

2. I do not think the solubility of the extra enzyme provided will influence the quality of the prep. To get a purer prep, you can wash the DNA one extra time. You can also use phenol-chloroform extraction before ethanol precipitation.
3. At the last step, remove the ethanol as much as possible, but do not dry the DNA too much. If it is too dry, it is difficult to dissolve. Also if you have a very high yield, you might want to increase the volume of TE to be able to resuspend everything.

Thank you for your enquiry and your interest in our products.

This kit ab65353 is a DNA Damage Quantification Kit which is to be used with purified DNA. You cannot use frozen tissues directly with this kit. The frozen tissue will need pre-processing to isolate the DNA. I would advise you to take a look at the on-line Protocol booklet (particularly Section 2 and Section 4) to get some further information of the recommended procedure.

I would suggest taking a look at an alternative kit ab65358 (Genomic DNA isolation kit) for this purpose.This productmay suit your need better:

I have a question regarding your DNA isolation kit. We have an experiment planned in which we want to separate the cytoplasmic DNA from the nuclear DNA. but we have a few questions regarding the usability for our experiment. We would like to know whether the cytoplasmic DNA is still usable for PCR after the use of your kit? I assume that this DNA ends up in the supernatant of step 3 of the protocol on your website. Thanks for your call last week and for your patience while I have been in touch with the production lab regarding ab65358. I apologize for the delay in getting back to you. This kit will isolate the total genomic DNA, however the minimum/ maximum lengths of fragmented DNA have not been experimentally determined. We don't have any data regarding this. I am sorry that we don't have further information, but if you have any other questions please let me know and I'll be happy to help.

Thank you for your inquiry. Yes, the customer should be able to use this kit. “For tissue samples, ground freshly excised tissue in liquid nitrogen. Weigh ~5 mg grounded fine tissue powder in a microcentrifuge tube.” The rest of the steps can be followed as indicated in the protocol linked on the datasheet. I hope this information helps. Thank you for your inquiry.

This kit has not been tested with dental plaque samples.

To my knowledge dental plaque consists mainly of bacteria.

Since this kit applied a extraction of the nucleus and then goes on to extract the DNA, it might be possible to extract the bacterial chromosome with the first lysis step. This has not been tested and optimized and I cannot recommend it for dental plaque.

I am sorry to confirm that we do not have any DNA isolation kit for bacteria in our catalog at the moment.
