Expression is up-regulated in cervical squamous cell carcinoma. Expressed in bone marrow cell line HS-27a which supports the long-term maintenance of immature progenitor cells. -

Defects in JAG1 are the cause of Alagille syndrome type 1 (ALGS1) [MIM:118450]. Alagille syndrome is an autosomal dominant multisystem disorder defined clinically by hepatic bile duct paucity and cholestasis in association with cardiac, skeletal, and ophthalmologic manifestations. There are characteristic facial features and less frequent clinical involvement of the renal and vascular systems.
Defects in JAG1 are a cause of tetralogy of Fallot (TOF) [MIM:187500]. TOF is a congenital heart anomaly which consists of pulmonary stenosis, ventricular septal defect, dextroposition of the aorta (aorta is on the right side instead of the left) and hypertrophy of the right ventricle. This condition results in a blue baby at birth due to inadequate oxygenation. Surgical correction is emergent. -

Differentiation of 3T3-L1 cells was performed by first growing cells to confluence (day 2) and then shifting them to differentiation medium after 48 h (day 0). The differentiation medium contained 1 μM dexamethasone, 0.5 mM IBMX, and 15μg/ml of insulin. Differentiation medium lacking dexamethasone and IBMX and addition of mouse TNF-alpha (20mg/ml) were both used as a control. As a negative control hCD137-Fc was used. Medium was changed to growth medium containing only insulin (15μg/ml) after 48 h (day 2). Staining with Oil Red O was typically performed on day 7. Notch signaling was generated by immobilizing recombinant mouse Jagged-1-Fc on plates (Jagged-1 was coated on 12-well plate at 5µg/ml (1µg/ml should also work) overnight at 4°C). These plates (with or without Notch ligand) were then used to grow 3T3-L1 cells.
