補足情報

TRAF7 also known as TNF receptor-associated factor 7 is an E3 ubiquitin ligase that plays a role in protein modification processes. It has a molecular mass of approximately 74 kDa. TRAF7 is ubiquitously expressed meaning it's found throughout various tissues in the body indicating its functional relevance across different biological processes. The presence of TRAF7 in diverse tissue types suggests its involvement in several cellular mechanisms although more research is necessary to fully understand its tissue-specific roles.
Biological function summary

The function of TRAF7 involves its action as part of the immune response and regulation of apoptosis. TRAF7 does not commonly form well-defined complexes but operates by tagging proteins with ubiquitin molecules marking them for degradation or altering their activity. This modification affects signal transduction pathways that govern cell growth and immune signaling highlighting TRAF7's role in maintaining cellular homeostasis and responding to external stimuli.

Pathways

TRAF7 participates in the NF-kB and JNK signaling pathways. These pathways play essential roles in inflammation immune response and stress response. TRAF7 interacts with other signaling proteins like TRAF2 and TRAF6 facilitating the propagation of signals that lead to activation of downstream transcription factors. These interactions emphasize TRAF7's significance in the balance of survival and apoptotic pathways impacting various cellular decisions and responses to environmental changes.

The dysregulation of TRAF7 has been associated with several conditions including cancer and genetic syndromes like meningioma a type of brain tumor. TRAF7 mutations often co-occur with mutations in genes associated with other pathways such as NF2 influencing tumorigenesis. Additionally abnormal TRAF7 function can affect immune responses potentially leading to autoimmune conditions by disturbing the normal signaling processes mediated by its interactions with proteins such as NF-κB. Understanding TRAF7's role can aid in developing therapeutic strategies for these disorders.

一般的な情報

機能

E3 ubiquitin and SUMO-protein ligase that plays a role in different biological processes such as innate immunity, inflammation or apoptosis (PubMed : 15001576, PubMed : 37086853). Potentiates MAP3K3-mediated activation of JUN/AP1 and DDIT3 transcriptional regulators (PubMed : 14743216). Negatively regulates MYB transcriptional activity by sequestering it to the cytosol via SUMOylation (By similarity). Plays a role in the phosphorylation of MAPK1 and/or MAPK3, probably via its interaction with MAP3K3. Negatively regulates RLR-mediated innate immunity by promoting 'Lys-48'-linked ubiquitination of TBK1 through its RING domain to inhibit the cellular antiviral response (PubMed : 37086853). Promotes 'Lys-29'-linked polyubiquitination of NEMO/IKBKG and RELA leading to targeting these two proteins to lysosomal degradative pathways, reducing the transcriptional activity of NF-kappa-B (PubMed : 21518757).
