    Inquiry:

    I intend to use this product to transform cells (EMT). I would like to know the recommended diluent for reconstitution (I thought to use the complete medium I use for the culture of the cells). If I used a sterile diluent, would it be necessary to sterile filter the solution, given that the vial is open in the laminar flow cabinet? Cells will be cultured with this protein for a few days. There are not specific instructions for reconstitution in the product manual.

    Below is the instructions for the reconstitution of this product:


    1. Centrifuge the vial prior to opening.

    2. Reconstitute with sterile water to a concentration of 0.1-1.0 mg/ml. Do not vortex.

    3. For extended storage, it is recommended to further dilute in a buffer containing a carrier protein (for example, 0.1% BSA) and store in working aliquots at -20oC to -80oC.

    The protein solution was sterile filtered through a 0.2 micron filter before lyophilization. However, you may filter the protein solution again to ensure the protein solution is sterile.

    I can confirm that the product can be reconstituted with sterile water at conc. 0.1-1mg/ml.

    Please note the concentration will depend on the amount of water you add. e.g. if 1ml is added the conc. will be 5ug/ml.

    Abcam product ab66045: anti TGF beta 2 antibody.
    Figure 2 of the product datasheet shows a Western blot for Anti-TGF-B2 antibody; yet, it does not indicate the species or tissue used. Also, the figure for the Western blot depicts one band at 13-kDa and another one at around 25 kDa; still, no indication which lysates from which species were analyzed, and which band is the actual one.

    The product description sheet indicates that Recombinant ab84070 is a homodimer with an expected molecular weight of 25kDa. The monomeric form is expected to run at approximately 12kDa. Is this information for ab84070 or ab66045?

    This information should be provided for ab66045. For this same figure, it is indicated that a secondary anti-goat antibody was used, yet ab66045 is an antibody made in rabbit.
    Moreover, in the product description sheet, it is indicated that the antibody detects a band of approximately 50 kDa in Western blots (tissue? Species?). Yet, in the image of the Western blot, there is not even a vestige of a band at 50 kDa.
    Please clarify.

    I am sorry that the caption describing the WB image was confusing. Please allow me to clarify:


    The image represents a WB in which the antibody ab66045 was used to detect the TGF beta 2 protein ab84070. Thus the sample was the recombinant protein and not a cell or tissue lysate. Both bands in the blot represent the target protein, one as a monomer and one as a dimer. The predicted molecular weight of the recombinantmonomer and dimer is 12kDa and 25kDa, respectively.


    Regarding endogenous protein, the predicted molecular weight is 50kDa, accounting for the discprepancy in size.


    Finally, regarding the secondary antibody, the text on the datasheet states: Goat polyclonal Secondary Antibody to Rabbit IgG - H&L (HRP), pre-adsorbed (ab97080). Thus, this secondary antibody was raised in goat, but is targeted to Rabbit IgG heavy and light chains.
