Tankyrase 2 also known as TNKS2 functions as a poly(ADP-ribose) polymerase adding ADP-ribose polymers to target proteins. It has a molecular weight of approximately 135 kDa. Expression of TNKS2 occurs in various tissues including the brain heart and skeletal muscle. This protein consists of several domains such as an ankyrin repeat domain and a sterile alpha motif (SAM) domain allowing it to interact with different substrates and mediate protein-protein interactions.
Biological function summary

TNKS2 plays a significant role in telomere maintenance and Wnt signaling. It is a component of the telomere-associated complex which helps regulate telomere length and stability by interacting with TRF1 a telomere-binding factor. In the context of Wnt signaling it functions by modulating the stability of Axin a scaffold protein thereby affecting the degradation of β-catenin. By ensuring proper telomere function and Wnt signaling TNKS2 supports cellular processes such as proliferation and differentiation.

Pathways

TNKS2 contributes notably to telomere maintenance and Wnt/β-catenin signaling pathways. In telomere maintenance its activity influences telomerase access to telomeres cooperating with proteins like TRF1 and TIN2. Within the Wnt/β-catenin signaling pathway TNKS2 interacts with Axin promoting β-catenin accumulation which in turn stimulates gene transcription necessary for cell growth and development. These two pathways demonstrate how TNKS2 is involved in regulating fundamental aspects of cell biology and growth.

TNKS2 associates with conditions such as cancer and fibrosis. In various cancers aberrant TNKS2 activity results in dysregulation of Wnt signaling leading to improper cell proliferation and tumor growth. For example its interaction with β-catenin contributes to oncogenesis in colorectal cancer. In fibrosis altered TNKS2 function disrupts normal signaling processes which can lead to the excessive connective tissue accumulation characteristic of fibrotic diseases. Therefore TNKS2 appears as a promising target in therapeutic strategies aiming to modulate these pathways and control disease progression.
