In many cases, microphthalmia/anophthalmia occurs in association with syndromes that include non-ocular abnormalities. MCOPS3 is characterized by the rare association of malformations including uni- or bilateral anophthalmia or microphthalmia, and esophageal atresia with trachoesophageal fistula.

Question

I have a question regarding the two products I bought from Abcam (ab95847 and ab79950)both SOX2 protein (Human). Can you please tell me the main differences between these to proteins except the fact that one is 20ug in a storage buffer and the other one is 5ug lyophilised? I would like to reassure you that our product have all been tested successfully and would be covered by our Abpromise guarantee.

Also, although we produce many in-house products, we obtain other products from a wide range of sources. In this case, these products are from different labs. Each product will be recombinant full length human SOX2. The only difference I noted beside those which you have mentioned is that ab79550 has > 95% purity by SDS-PAGE whereas ab95847 has > 90% purity by SDS-PAGE.

I am still in the process of searching for high affinity antibodies for SOX2 protein I bought from Abcam (ab95847). Based on what we talked before, you recommended two monoclonal Abs (ab75485, and ab95331) plus a polyclonal (ab75627) against the Sox2 Ag(ab95847). I was wondering if any other higher quality SOX2 Abs might have been available by Abcam or you would still recommend the above mentioned products. Also, I have received a coupon by Abcam, "4 for 3". What other high affinity SOX2 Ab would you recommend (100ug, ~1 mg/ml) so that I could benefit from the offer. I found the following Abs in your website (ab129031) and (ab130225), but not sure whether these monoclonals recognize a different epitope than the other monoclonals you recommended before (mentioned above). Just a reminder that I need to have a high affinity pair of monoclonal or a very good polyclonal for doing an immunoassay type of experiment (sensitivity (limit of detection) is extremely important for my application). Also, I remember you told me over the phone that the SOX2 Ag (ab79950) might be a better option than the Ag (ab95847) I have already bought from your company. Please let me know if the Abs you recommended before would have a higher affinity for this Ag as opposed to the Ag I already have, as if that is the case, then I can use the coupon to buy this Ag as my 4th product if no other high quality Ab is available for my research. Please advise and thank you very much in advance for your time and support.

While I do not know exactly what you and John spoke of on the phone, from you email I have gathered that you are looking for very high affinity Sox2 antibodies for used in you sensitivity assay. Generally we do not have kD values for products on our catalog. However, for higher affinity products I would recommend use of antibodies raised in rabbit, especially rabbit monoclonals.The mechanisms of antibody affinity maturation employed by rabbits allow them to generate antibodies with very high affinity and specificity. Typically rabbit monoclonal antibodies (RabMAbs) have 10 - 100 times higher affinity than mouse monoclonal antibodies.

This product has been tested in sandwich ELISA with the rabbit monoclonal ab92494. These tests also included the use of Sox2 ag ab79950. From your email I can see that my colleague did recommend this. It seems likely that his recommendation was in part based on this data.
