Biological function summary
SIT plays an important role in regulating T-cell receptor (TCR) signaling. By participating in the assembly of signaling complexes it modulates the strength and duration of TCR signals. SIT does not act alone but often interacts with other adaptor proteins such as LAT (Linker of Activated T cells) and GRB2 (Growth Factor Receptor-Bound Protein 2). This interaction framework contributes to the modulation of downstream cellular responses affecting activation and differentiation within the T-cell population.
Pathways
SIT engages heavily in the TCR signaling pathway. In this pathway SIT works alongside LAT and another protein called ZAP-70 (Zeta-chain-associated protein kinase 70) to propagate key signaling events. SIT's role in signal transduction implies it indirectly influences the MAPK/ERK pathway a critical route for cell proliferation differentiation and apoptosis. Through these pathways SIT ensures that T-cells respond appropriately to antigens maintaining immune system homeostasis.
