補足情報

Haptoglobulin beta also known as Hp beta is a component of the haptoglobulin protein which binds free hemoglobin released from erythrocytes. This prevents oxidative damage to tissues. Haptoglobulin has a molecular mass of approximately 40 to 45 kDa depending on glycosylation. It is mainly expressed in the liver and then secreted into the bloodstream acting as an acute-phase protein during inflammatory responses.
Biological function summary

Haptoglobulin beta plays an important role in hemoglobin transport by forming a stable complex with free hemoglobin therefore facilitating its clearance by macrophages. This protein is a part of the haptoglobin-hemoglobin complex which is essential for preventing kidney damage and conserving iron after red blood cell lysis. Its biological function highlights its significance in the body's defense against oxidative stress.

Pathways

Haptoglobulin beta is involved in iron recycling and inflammation regulation. The hemoglobin scavenger receptor CD163 mediates the uptake of the haptoglobin-hemoglobin complex playing an important role in the iron metabolism pathway and immune system functioning. Additionally haptoglobulin is connected to the acute-phase response pathway alongside proteins like serum amyloid A to moderate inflammation and tissue protection.

Haptoglobulin beta links to hemolytic anemia and cardiovascular disease. A deficiency or polymorphism in the haptoglobulin gene can exacerbate complications in these conditions due to impaired hemoglobin scavenging. Heme released in absence of effective haptoglobulin activity promotes oxidative damage and inflammation contributing to disease pathology. C-reactive protein often interacts with haptoglobulin in the inflammatory network highlighting its multifaceted role in disease processes.

一般的な情報

機能

As a result of hemolysis, hemoglobin is found to accumulate in the kidney and is secreted in the urine. Haptoglobin captures, and combines with free plasma hemoglobin to allow hepatic recycling of heme iron and to prevent kidney damage. Haptoglobin also acts as an antioxidant, has antibacterial activity, and plays a role in modulating many aspects of the acute phase response. Hemoglobin/haptoglobin complexes are rapidly cleared by the macrophage CD163 scavenger receptor expressed on the surface of liver Kupfer cells through an endocytic lysosomal degradation pathway.. The uncleaved form of allele alpha-2 (2-2), known as zonulin, plays a role in intestinal permeability, allowing intercellular tight junction disassembly, and controlling the equilibrium between tolerance and immunity to non-self antigens.

配列の類似性

Belongs to the peptidase S1 family.
