Biological function summary
The NS1 protein impacts several cellular processes within the host. NS1 is not part of a larger protein complex but functions significantly on its own to facilitate a favorable environment for the virus. It assists in immune evasion by modulating host immune signaling pathways and inhibiting complement activation. NS1 can also bind to and disrupt endothelial cells which contributes to vascular leakage a severe clinical symptom associated with dengue virus infection.
Pathways
NS1 protein interacts with several pathways in its pursuit of viral propagation. It engages notably with the unfolded protein response (UPR) and the Toll-like receptor (TLR) pathways. Within the UPR NS1 modulates stress responses to maintain cellular homeostasis under viral replication conditions. In the context of the TLR pathway NS1 is recognized by TLR4 leading to the production of inflammatory cytokines. NS1's interactions within these pathways are important for understanding how dengue virus evades host immune responses.
