      Anti-VDAC/Porin Antibody is directed against human VDAC1/Porin1 protein. A BLAST analysis was used to suggest that this antibody would react with VDAC1/Porin1 from a wide range or organisms, including avian, mammalian, aquatic and reptilian sources based on 100% homology for the immunogen sequence. Cross reactivity will occur with all forms of VDACs including VDAC1, VDAC2 (4 isoforms) and VDAC3 (2 isoforms). Such broad reactivity makes this antibody useful as an excellent loading control (mitochondrial).

      Forms a channel through the mitochondrial outer membrane and also the plasma membrane. The channel at the outer mitochondrial membrane allows diffusion of small hydrophilic molecules; in the plasma membrane it is involved in cell volume regulation and apoptosis. It adopts an open conformation at low or zero membrane potential and a closed conformation at potentials above 30-40 mV. The open state has a weak anion selectivity whereas the closed state is cation-selective. May participate in the formation of the permeability transition pore complex (PTPC) responsible for the release of mitochondrial products that triggers apoptosis.

      Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-VDAC1/Porin antibody (ab34726)
      IHC image of ab34726 staining in human liver formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab34726, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

      For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

    Hi, The technical team agreed to send a replacement antibody free of charge after the VDAC antibody (ab34726 lot: GR95237-1) we purchased failed to perform as it should. I was given the order number 1164302 for the replacement and would like to know what is the status of this order please, as I have not yet received the item,

    I have issued a free of charge replacement with a different lot number, but it looks like that this vial was already on the pick list for another order. Therefore, the order is waiting to receive a vial we already have been sold and can't be fulfilled. Unfortunately, this has not been flagged up... I prologize for the delay!

    Checking our stock, it looks we do not have an alternative lot. However, your complaint has been the only one so far this year. Therefore, I do not think that we have a general lot problem, but that you unfortunately, received a faulty vial. Your free of charge replacementwould be covered as well with our guarantee, so I would like to encourage you to try the same lot again.

    If you would prefer instead another antibody against this target, a credit note or a refund, I am happy to issue this for you as well.

    Thank you for your reply but I think perhaps I was not very clear in my
    email - we used the antibody at 1/500 but were still unable to get any
    bands. I have attached a scan (a) of a blot using ab34726 at 1/500 where
    30ug of protein was loaded.
    Lane 1= rat liver mitochondria
    Lane 2= digitonin treated mitochondria (mitoplasts)
    Lane 3= supernatant
    Lane 4= digitonin treated mitochondria
    Lane 5= supernatant
    We would expect a strong signal, especially in Lane 1 but there is nothing.
    The same membrane was then stripped and reprobed with an alternative VDAC
    antibody (monoclonal) (b) and as you can see we then obtained bands.
    I really do feel the latest batch of your VDAC antibody (ab34726) is not
    performing as it should, so would again request a replacement batch please.

    We have used the ab34726 VDAC antibody for Western blotting on rat heart
    mitochondria in our lab for several years and have always found it to be a
    very reliable antibody until we purchased a new batch on 14 August 2012
    (Purchase Order FMVS1176576), which despite several attempts on several
    preparations of mitochondria and using the antibody at a dilution as low as
    1/500 (where previously it has worked fine at 1/3000) have failed to get
    any decent bands around the 32kDa area (or at any other molecular weight!).
    The antibody we were sent is: LOT GR95237-1. Have you had any other
    customers reporting problems with this? Can you please send us a
    replacement antibody of a different lot number as we know this antibody
    usually works well and can only assume there is something not quite right
    with the batch we were sent?

    Although other lots of this antibody have worked well under these former conditions, all individual antibodies will require individual optimization.

    According to the datasheet, 1/500 is within the recommended dilution range. It may be that this lot has a lower affinity asthe previous ones and therefore need a higher concentration. As the antibody is workingwithin the givenparameters,I am afraid I cannot send out a replacement

    Our customer would like to use ab14734 for IHC-P, staining of human cancer sections (from different origins) and healthy human sections. Kindly advise if this specific antibody (ab14734) will help to distinguish between healthy and cancer tissue? In addition to that, please explain the difference between  Anti-VDAC1 / Porin antibodies with the addition of "Mitochondrial Loading Control" after the name of the antibody (like ab14734) and Anti-VDAC1 / Porin antibodies that lack this description (ab34726, for ex).     Thank you for your inquiry. VDAC1 is a mitochondrial protein and I believe its level of expression is pretty ubiquitous in all tissues, cancerous and normal. According to the Human Protein Atlas, VDAC1 is expressed in most normal and cancer tissues tested. http://www.proteinatlas.org/ENSG00000213585/normal http://www.proteinatlas.org/ENSG00000213585/cancer All anti-VDAC1/Porin antibodies in our catalogue will stain the same mitochondrial protein. The ones named "Mitochondrial Loading Control" have typically been cited in papers to have been used as such. That is really the only main difference in the name. I hope this information helps.
