      Cell adhesion protein that participates in lymphocyte recirculation by mediating the binding of lymphocytes to peripheral lymph node vascular endothelial cells in an L-selectin-independent fashion. Has a monoamine oxidase activity. May play a role in adipogenesis.

      please find here below our customer's answer:
      Tumor tissues are about 1cm in diameter, therefore the entire tissue had to be fixed for at least 2 hs, then it was incubated in sucrose 10%-20%-30% in order to preserve the tissue from the freezing. Incubation time depends on the tissue: when the sucrose solution spreads into the entire tissue, the formalin is washed away and the tissue moves from the top to the bottom of the solution: after this, the solution can be changed. Tissue was briefly fixed again (10 minutes, in the same solution of 10% formalin buffered), before the application of labelling protocol. This is a standard protocol to fix a tissue; it is possible that the tissue results over-fixed, but we never had problems to highlight a marker with an antibody. It is possible that this particular antigen requires an antigen retrieval procedure in addition to the standard labelling protocol. Could you suggest me some antigen retrieval protocol?

      Answer

      Thank you for having followed this up with the customer.

      Indeed, the protocol seems to me to be intermediate in the fixation with paraffin embedded tissues, where the tissues are often fixed overnight in PFA, and with frozen sections, where the sections are just postfixed.
      We do have unfortunately no experience with this antibody on paraffin embedded orotherlong term PFA fixed tissue(such aswholemount)and I can not recommend on particular antigen retrieval method. I have not found a reference in this regards in the literature neither.
      I can not guarantee that this will work, however there is neverthelessa good chance that antigen retrieval couldallow antibody staining.I would therefore recommend the customer to test different antigen retrievals, such as heat induced antigen retrieval with low and high pH (e.g. with Citrate and EDTA buffers). Here is publication comparing different antigen retrievals- Emoto et al., J Histochem Cytochem. 2005 Nov;53(11):1311-21

      I hope this makes sense. Please do let me know how the customer is proceeding. Ifthe antigen retrieval will not allow good results, I will arrange for a replacement or credit.

      It states that the sections have been prefixed in formalin. Can you please provide more information on this step? Could it be that the samples have been overfixed and an antigen retrieval would be necessary?
