USP26 also known as Ubiquitin Specific Peptidase 26 is an enzyme that removes ubiquitin moieties from specific proteins regulating their degradation localization or activity. USP26 is part of the deubiquitinating enzymes (DUBs) family and has a molecular mass of approximately 104 kDa. It expresses mainly in testicular tissue which indicates its role in reproductive biology.
Biological function summary

The enzyme plays a role in spermatogenesis the process of sperm cell development. It participates in the regulation of the androgen receptor (AR) signaling pathway. By interacting with receptors it stabilizes and modulates their activity contributing to normal testicular function. This connects USP26 to a protein complex involving androgen receptors which are essential for male reproductive health.

Pathways

The deubiquitinating activity of USP26 influences the androgen receptor signaling pathway important for male reproductive processes. In the ubiquitin-proteasome system USP26 modulates the stability of the AR receptor and other proteins related to this pathway which may include factors like HSP90 that aid in receptor activation and stabilization.

Anomalies in USP26 function correlate with male infertility. Mutations or altered expression are linked to disorders in spermatogenesis. This has connections with androgen receptor abnormalities as proper receptor function is necessary for normal sperm production. Studies also find indications that dysregulation may link to prostate cancer where androgen receptor involvement is critical.

Involved in somatic cell reprogramming through the 'Lys-48'-linked deubiquitination and stabilization of CBX4 and CBX6, two components of the polycomb-repressive complex 1 (PRC1) (PubMed : 28839133). Also deubiquitinates and probably stabilizes the androgen receptor (AR), regulating the androgen receptor signaling pathway (PubMed : 20501646).
