機能
Ligand for the NKG2D receptor, together with at least ULBP1 and ULBP3. ULBPs activate multiple signaling pathways in primary NK cells, resulting in the production of cytokines and chemokines. Binding of ULBPs ligands to NKG2D induces calcium mobilization and activation of the JAK2, STAT5, ERK and PI3K kinase/Akt signal transduction pathway. In CMV infected cells, interacts with soluble CMV glycoprotein UL16. The interaction with UL16 blocked the interaction with the NKG2D receptor, providing a mechanism by which CMV infected cells might escape the immune system. UL16 also causes ULBP2 to be retained in the ER and cis-Golgi apparatus so that it does not reach the cell surface. -

Flow Cytometry analysis of Raji (Human Burkitt's lymphoma B lymphocyte, Left) / BxPC-3 (Human pancreas adenocarcinoma epithelial cell, Right) labeling ULBP2 (red) with ab290731 at a 1/500 dilution. A Goat Anti-Rabbit IgG (Alexa Fluor® 488, ab150081) was used as the secondary antibody at a 1/2000 dilution. Black - Isotype control: Rabbit monoclonal IgG (ab172730). Blue - unlabeled control: Cells without incubation with primary and secondary antibodies.
Negative control: Raji.
Gated on viable cells.

Intracellular Flow Cytometry analysis of HEK-293T (Human embryonic kidney epithelial cell) transfected with a human ULBP2 expression vector labeling ULBP2 with ab290731 at a 1/500 dilution (Right). Cells were fixed with 2% paraformaldehyde and permeabilized with 0.1% Tween-20. A Goat Anti-Rabbit IgG (Alexa Fluor® 488, ab150081) was used as the secondary antibody at a 1/2000 dilution. Isotype control: Rabbit monoclonal IgG (ab172730) (Left).
Cells were surface stained with rabbit IgG or ab290731, then fixed with 2% PFA followed by intracellular staining with Myc-tag conjugated to Alexa Fluor® 647.
