TTLL5 or Tubulin Tyrosine Ligase Like 5 acts as an enzyme that adds glutamates onto tubulin a process known as polyglutamylation. It has an approximate mass of 121 kDa. This protein is expressed in various tissues with higher levels observed in the retinal photoreceptor cells and testes. TTLL5 plays a role in the post-translational modification of proteins which is essential for the functioning of the microtubule network.
Biological function summary

TTLL5 contributes to cellular processes by modifying tubulin a critical component of the cytoskeleton. In its role it performs polyglutamylation affecting microtubule stability and interactions with microtubule-associated proteins (MAPs) and motor proteins. TTLL5 functions as a part of a larger group of enzymes similar to other members of the tubulin tyrosine ligase-like family which regulates the different aspects of microtubule dynamics.

Pathways

TTLL5 is important in the MAPK signaling pathway and axon guidance. In these pathways TTLL5 affects cellular morphology and signaling by modifying tubulin structures which may regulate the activity of proteins like MAP2 and tau impacting cell transport and division. Its role in these pathways highlights the significance of microtubule modifications in both cellular signaling and structural integrity.

Mutations or dysregulation of TTLL5 have been linked to retinal degeneration and ciliary dysfunction disorders. In retinal degeneration TTLL5 defects affect photoreceptor cell functions due to improper protein modification. The protein’s impact on the disease connects through interaction with other microtubule-related proteins which are also critical for maintaining retinal cell structure and function.

Polyglutamylase which modifies tubulin, generating polyglutamate side chains on the gamma-carboxyl group of specific glutamate residues within the C-terminal tail of tubulin. Preferentially mediates ATP-dependent initiation step of the polyglutamylation reaction over the elongation step. Preferentially modifies the alpha-tubulin tail over a beta-tail (By similarity). Required for CCSAP localization to both polyglutamylated spindle and cilia microtubules (PubMed : 22493317). Increases the effects of transcriptional coactivator NCOA2/TIF2 in glucocorticoid receptor-mediated repression and induction and in androgen receptor-mediated induction (PubMed : 17116691).
