Anti-TSLP 抗体 (ab115700)

I now have further information on how the IHC staining of the mouse liver and spleen tissue presented on the datasheet for anti-TSLP antibody ab115700. The protocol was as follows:


1. Formlin fixation


The tissue fixed in 10% neutral buffered formalin at room temperature for approximately 16 hours then embedded in paraffin wax


2. Tissue sectioning and deparaffinization


4 um sections were cut and fixed to microscope slides then heated in a tissue-drying oven for 45 minutes at 60°C. Deparaffinization and rehydration steps were then performed.


3. Antigen retrieval


The tissue was treated with 0.01 Msodium citrate buffer, pH 6.0 at 99-100°C for 20 minutes. The tissue was then allowed to cool and rinsed in TBST for 1 minute at room temperature.


4. Immunostaining


The tissue was blocked with universal protein block for 20 minutes at room temperature then incubated with the primary antibody (5 ug/mL in universal protein block) for 45 minutes at room temperature. The slide was then rinsed with TBST and the secondary antibody applied for 30 minutes at room temperature.
