  • 機能

    Receptor-activated non-selective calcium permeant cation channel involved in detection of noxious chemical and thermal stimuli. Seems to mediate proton influx and may be involved in intracellular acidosis in nociceptive neurons. May be involved in mediation of inflammatory pain and hyperalgesia. Sensitized by a phosphatidylinositol second messenger system activated by receptor tyrosine kinases, which involves PKC isozymes and PCL. Acts as ionotropic endocannabinoid receptor with central neuromodulatory effects. Triggers a form of long-term depression (TRPV1-LTD) mediated by the endocannabinoid anandamine in the hippocampus and nucleus accubens by affecting AMPA receptors endocytosis.

    Widely expressed at low levels. Expression is elevated in dorsal root ganglia. In skin, expressed in cutaneous sensory nerve fibers, mast cells, epidermal keratinocytes, dermal blood vessels, the inner root sheet and the infundibulum of hair follicles, differentiated sebocytes, sweat gland ducts, and the secretory portion of eccrine sweat glands (at protein level).

Dephosphorylation by calcineurin seems to lead to receptor desensitization and phosphorylation by CAMKII recovers activity.

    Question

    I tried to test the indicate protocol : https://www.abcam.com/VR1-antibody-ab3487.html&tab=abreviews&intabreviewid=26192 : test fixation methanol, over night, goat serum saturation, permeabilization triton X100

    But without success !

    To answer to your question (Can you confirm that the other reactives (e.g. the secondary antibody) do work fine when used with other primary rabbit antibodies?) , yes, we use this secondary antibody frequently and it work very well.

    Please can you give me a solution, my client want a VR1 labelling on human skin !

    Inquiry: Mr or Mrs I tried the antibody ab3487 (anti-VR1) on frozen section of skin : - fixed in aceton/methanol - saturation in milk - antibody dilution : 1/1000 and 1/100, 1 hour at RT - secondary antibody GAR alexa fluor 488, 1 hour at RT - wash in PBS-tween 20. It didn't work. Do you have a specific protocol for this marker ? Could you give me the conditions please ? Thanks a lot. Regards

    Thank you for contacting us. I am sorry to hear that this antibody is not providing satisfactory results.

    We do not have a specific protocol for this antibody. We expect this antibody to work with a general IHC protocol after the classic optimisation steps. Looking at the information you provided, I would like to offerthe followingsuggestions to help optimize the results from ab3487. I would also appreciate if you can confirm some further details:

    1.) Can you please confirm of which species the skin sections usedare coming from. We have tested this antibody so far only on human tissue. It might reacts with other species, however we need to test this first.

    2.) I can recommend to incubate the antibody overnight at 4°C. This can increase the staining intensity.

    3.) I can also recommend to test different blocking solutions. This can significantly improve the staining. I can recommend to test BSA and serum instead of milk. As serum you would use serum of the species of the secondary antibody (goat?). The researcher having submitted the feedback on this antibody, has used for example 10% serum.     4.) Can you confirm that the other reactives (e.g. the secondary antibody) do work fine when used with other primary rabbit antibodies?

    5.) Have you tested other fixation methods as well, such as Methanol only or Paraformaldehyde? The user mentionned above, had used succesfully Methanol only, whereas the lab has tested the antibody also in paraformaldhyde fixation obtaining good results.

    I hope these suggestions will help to identify the problem. Should the suggestions not improve the results, please do let me know.
