TRPM7 and TRPM6 are members of the transient receptor potential melastatin subfamily often referred to as TRP-PLIK or CHAK1 for TRPM7. TRPM7 has a molecular mass of approximately 165 kDa while TRPM6 is roughly 230 kDa. These proteins function as ion channels with kinase activity. They regulate the homeostasis of divalent cations like magnesium and calcium by permitting their entry into cells. TRPM7 is expressed in various tissues including the heart liver and brain while TRPM6 is mostly found in the kidney and intestine.
Biological function summary

TRPM7 and TRPM6 play essential roles in cellular magnesium absorption and homeostasis. They form homo- or heteromeric channel complexes with the combination of TRPM6 and TRPM7 enhancing channel function. These proteins influence critical cellular processes such as proliferation apoptosis and migration by modulating cellular magnesium levels. TRPM7's distinct kinase domain also impacts intracellular signaling pathways through phosphorylation of specific substrates regulating response to cellular stress and differentiation.

Pathways

TRPM7 and TRPM6 modulate cell signaling pathways linked to magnesium-sensitive enzymes and proteins like those involved in the PI3K/Akt signaling pathway. This pathway is integral for cell growth survival and metabolism. TRPM7 interacts with various signaling proteins including PLC and MAPK influencing cellular responses to external stimuli. TRPM6 often partners with TRPM7 to fine-tune these cellular pathways maintaining appropriate ionic balances necessary for cellular functions.

TRPM7 and TRPM6 are linked to magnesium homeostasis-related conditions such as hypomagnesemia and hypertension. Variants in TRPM6 associate with hypomagnesemia with secondary hypocalcemia a genetic disorder affecting magnesium uptake in the intestine. Bifunctional protein that combines an ion channel with an intrinsic kinase domain, enabling it to modulate cellular functions either by conducting ions through the pore or by phosphorylating downstream proteins via its kinase domain (PubMed : 14576148, PubMed : 16636202, PubMed : 18258429, PubMed : 18365021). Crucial for Mg(2+) homeostasis. Has an important role in epithelial Mg(2+) transport and in the active Mg(2+) absorption in the gut and kidney (PubMed : 14576148). However, whether TRPM6 forms functional homomeric channels by itself or functions primarily as a subunit of heteromeric TRPM6-TRPM7 channels, is still under debate (PubMed : 14576148, PubMed : 16636202, PubMed : 24385424).. TRPM6 kinase, cleaved form. The C-terminal kinase domain can be cleaved from the channel segment in a cell-type-specific fashion.
