Western blot - Anti-TRIM33 antibody [EPR25102-19] (BSA and Azide free) (AB300147)
This data was developed using ab300146, the same antibody clone in a different buffer formulation.
Blocking and diluting buffer and concentration : 5% NFDM/TBST.
Lanes 1-4 : Merged signal (red and green). Green - ab300146 observed at 150 kDa. Red - loading control ab8245 observed at 36 kDa.
ab300146 Anti-TRIM33 antibody [EPR25102-19] was shown to specifically react with TRIM33 in wild-type HAP1 cells. Loss of signal was observed when knockout cell line was used. Wild-type and TRIM33 knockout samples were subjected to SDS-PAGE. ab300146 and Anti-GAPDH antibody [6C5] - Loading Control (ab8245) were incubated at 4℃ overnight at 1 in 1000 dilution and 1 in 20000 dilution respectively.
Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preadsorbed (ab216773) and Goat anti-Mouse IgG H&L (IRDye® 680RD) preadsorbed (ab216776) secondary antibodies at 1 in 10000 dilution for 1 hour at room temperature before imaging.

293T (human embryonic kidney epithelial cell) whole cell lysate

Biological function summary
The ubiquitination actions of TRIM33 modulate transcriptional regulation and cell fate decisions. TRIM33 interacts with transcription factors and chromatin often forming part of the larger transcriptional regulatory complexes. Its role in repressing TGF-β signaling by ubiquitylating SMAD proteins highlights its influence on cellular growth and differentiation.
Pathways
TRIM33 has an impact on the TGF-β and BMP signaling pathways both essential for controlling cell proliferation and apoptosis. It acts in partnership with other proteins such as SMAD4 to negatively regulate TGF-β responses helping balance cellular growth.
