I am a little baffled as to why you are still not getting a signal, but I do not think that it is you, its probably either something really simple that we have overlooked, or its bad luck that we sent you two antibodies that are not working for you.

Have you successfully used these samples in conjunction with any other primary anybody (eg actin, tubulin etc, some other housekeeping protein?). Also, after you transfer you samples to the nitrocellulose or PVDF membrane, do you do a Poncea stain to check that the transfer worked correctly? What is the longest exposure that you do of the membrane after you have added the ECL?

We will figure this one out and hopefully it’s not the case that we have sent you 2 antibodies that are not working for you.

I am really sorry about the issues that you are having. I think the best way forward is for me to send you ab67192 and a new vial of the spleen lysate (it should work as a positive control for ab67192. I would send both of these free of charge, since the original antibody is not working and the fact that you have used all of the lysate trying to get it to work.

We do not have small/sample sizes of our antibody, so I am unable to send a small volume of ab83890 and also we do not have a different lot of it in stock.

If this sounds like a good plan to you, then let me know and I will have them sent to you as soon as possible. Also I think I have found your order for these products, but to confirm that I have the correct on, could you please confirm the name of the PI that you work for.

I understand your hesitation to purchase a new antibody and you could try and use 5%BSA as a blocking reagent, but the fact that you are seeing no signal what so ever makes me think that changing the block may not be successful, but it is worth a go.

Since the first antibody did not work for you, I could send ab67192 as a free of charge replacement to you. You would not be charged anything for it. Or I could process a refund for the cost of ab83890.

I think the best way to move forward from here is to either replace the current antibody you have with ab67192:

Or, I can refund the cost of the original purchase. I do not think that there is any other protocol tips that I could suggest that would get the current antibody to produce a signal. Unfortunately there must just be something wrong with that particular vial of antibody.

I am sorry about the issues you have been having with ab83890 in western blot. The antibody is covered under our Abpromise for six months and is guaranteed to work in western blot on human samples. I have checked with the lab and they sent me the following information:

The antibody was tested in mouse brain and shows a band at the same size as the human spleen image on the datasheet. We have only tested the antibody in IHC-Pusing human spleen tissue sections (please see image attached). The antibody has not been tested with knockout mice.
