TOX2 also known as Thymocyte Selection-Associated High Mobility Group Box Protein 2 is a transcription factor weighing approximately 53 kDa. It is a nuclear protein expressed mainly in the T-cells of the thymus and spleen with notable expression in the brain and bone marrow. TOX2 belongs to the TOX family of proteins characterized by the presence of a high-mobility group (HMG) domain which facilitates its ability to bind and bend DNA thereby influencing the transcription of target genes.
Biological function summary

TOX2 plays an important role in the development and function of the immune system. It assists in the maturation of T-cells and the differentiation of thymocytes being part of a complex transcriptional network. TOX2 interacts with other transcription factors to modulate gene expression profiles critical for immune responses. It has implications in regulating aspects of inflammation and peripheral tolerance indicating its influence on immune homeostasis.

Pathways

TOX2 integrates into processes that govern T-cell receptor signaling and thymocyte development. It associates with pathways such as the Notch signaling pathway influencing thymocyte lineage decisions. Additionally TOX2 interacts with proteins like GATA3 and RUNX1 which are involved in the regulation of lymphoid cell development and differentiation further emphasizing its role in fine-tuning T-cell development and function.

Alterations in TOX2 expression or function bear connections to autoimmune conditions and certain cancers.
