      ICC/IF image of ab57523 stained HeLa cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab57523, 10µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-mouse IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

      Overlay histogram showing HeLa cells stained with ab57523 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab57523, 1µg/1x106 cells) for 30 min at 22°C. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22°C. Isotype control antibody (black line) was mouse IgG2a [ICIGG2A] (ab91361, 1µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed.

    - ab57523 giving 2 prominent bands at 30kDa and 88 kDa.
    - Used TOMM22 as marker for outer mitochondrial membrane proteins
    - What is the identity of 88 kDa band?

    Thank you very much for your patience while I was in discussion with lab.

    My colleague has confirmed that, they tested ab57523 with PC-12, Raw 264.7 and NIH/3T3 cells in western blot and they haven't observed any band at 88kDa. The band they observed was near ˜30kDa.

    For ab48139 they suggested the lower bands might be degradation product or uncharacterized proteins that cross-react with the antibody.

    I am sorry I am unable to find a more definite explanation of the bands you have been observing. We have also received no complaints for these antibodies so protocol troubleshooting might help in this case. Please try tweaking the dilution factor, lysate concentration etc.

    It appears the protein name in Yeasts is TOM22 and not TOMM22. Below is it's DNA sequence. The currently selected gene/sequence is : TOM22/YNL131W on chromosome XIV from coordinates 378767 to 379225. This is the genomic DNA sequence and may contain untranslated regions or bases (e. g. introns, or the site of a programmed translational frameshift). For a version of the sequence that does not contain these untranslated sequence region(s), please go back and select the coding sequence option.     Thank you for providing this extra information. The alignment shows a very low homology between the yeast protein and the human TOMM2 protein (www.uniprot.org/uniprot/Q9NS69), 16% only. This result suggests that unfortunately the TOMM2 antibody ab57523 is not suitable for the detection of the yeast protein. I hope this information is nevertheless useful for you.     Thank you very much for your inquiry. Unfortunately we do not have an antibody in our catalogue for the moment for which we can suggest a crossreactivity with the yeast SEC61 proteins. I am very sorry for this. Please see further below for the alignment scores. For the TOMM22 protein, I could not find the sequence in UniProt neither. Can the customer maybe provide this?
