      Cytokine that binds to TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. It is mainly secreted by macrophages and can induce cell death of certain tumor cell lines. It is potent pyrogen causing fever by direct action or by stimulation of interleukin-1 secretion and is implicated in the induction of cachexia, Under certain conditions it can stimulate cell proliferation and induce cell differentiation.

      Genetic variations in TNF are a cause of susceptibility psoriatic arthritis (PSORAS) [MIM:607507]. PSORAS is an inflammatory, seronegative arthritis associated with psoriasis. It is a heterogeneous disorder ranging from a mild, non-destructive disease to a severe, progressive, erosive arthropathy. Five types of psoriatic arthritis have been defined: asymmetrical oligoarthritis characterized by primary involvement of the small joints of the fingers or toes; asymmetrical arthritis which involves the joints of the extremities; symmetrical polyarthritis characterized by a rheumatoidlike pattern that can involve hands, wrists, ankles, and feet; arthritis mutilans, which is a rare but deforming and destructive condition; arthritis of the sacroiliac joints and spine (psoriatic spondylitis).

    Hello, We are developping a double elispot IFNg/TNFa in mouse. We already have an IFNg elispot with biotin / streptavidin / HRP detection system, so we are looking for antibodies for the TNFa part. I have seen these antibodies :- ab11564 for coating- ab34674 for detection- ab6113 for detection anti species What do you think of this combination?Thanks a lot

    Thank you very much for your inquiry. Reviewing the datasheets for these antibody I am confident that a single ELISPOT should give good results with this combination of antibodies. But unfortunately, these antibodies have not been tested in ELISPOT (only ELISA) and are therefore not covered by our Abpromise guarantee. I do not have experience with double ELISPOTs and therefore do not want to make a prediction. You might be eligible for our testing discount program if you would also perform a single ELISPOT with these antibodies. Please let me know if you are interested in this offer.     Thanks for your response. So what is this integral membrane protein? Actually, the band I am getting is close to 25 kDa. So this is not TNF-alpha? Please tell me more about this integral membrane protein. I thought it is TNF-alpha because it is really a very clean band.     Thank you for your email. Our source for this antibody tested ab11564 in Western blotting with recombinant TNF alpha. Conditions were standard. To use this antibody in Western blotting, I would recommend referring to our general Western blot protocol which can be found in the protocol section of the online datasheet. I would suggest using ab11564 at a 1:1000 dilution, and ab6734 at a 1:5000 dilution. I conferred with our source for ab11564 and it sounds like you are detecting the membrane bound form of TNF alpha (the 26 kDa integral membrane protein).     This antibody recognises the 17 kDa soluble form of TNF alpha. TNF alpha forms dimers and trimers, and in addition is found as a 26 kDa integral membrane protein. I am not able to comment on the ability of this antibody to recognise the membrane bound form but I can tell you that we also see a trimer of the soluble form in WBs (running at around 50 kDa). Can you tell me whether this information is up-to-date please? Also, would reactivity with TNF beta be likely? Are the amino acid sequences for TNF alpha and TNF beta similar?

    Thank you for your enquiry and for your patience. All the information that we have regarding these two antibodies is loacted on the online datasheets. To our knowledge, reactivity with TNF beta has still not yet been tested. Mature TNF-beta and TNF-alpha share approximately 35% protein sequence homology, and the biologically active secreted forms of both proteins are trimers.
