Toll like receptor 4 (TLR4) is a member of the Toll like receptors (TLRs) family comprising a group of evolutionary conserved pattern recognition molecules that play a key role in host defense during microbial infection by regulating both innate and adaptive immune responses. TLRs are type I transmembrane proteins with ectodomains containing interspersed leucine rich repeat (LRR) motifs that are involved in the recognition of various microbial ligands. All members of the family share a sequence in their cytoplasmic domain with the Type I IL1 receptor (TIR) and this highly conserved motif is involved in bimolecular interactions necessary for signal transduction. Cooperates with MD2 and CD14 to mediate the innate immune response to bacterial lipopolysaccharide (LPS). Acts via MYD88, TIRAP and TRAF6, leading to NF-kappa-B activation, cytokine secretion and the inflammatory response. Also involved in LPS-independent inflammatory responses triggered by Ni2+. These responses require non-conserved histidines and are, therefore, species-specific.
