Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-Sulfatase 1/SULF1 antibody (AB32763)
IHC image of Sulfatase 1/SULF1 staining in Human skeletal muscle formalin-fixed paraffin embedded tissue section*, performed on a Leica Bond™ system.. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab32763, 0.1 μg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

Biological function summary
SULF1 plays a significant role in modulating cellular processes like growth angiogenesis and tissue repair by altering the bioavailability of heparan sulfate-bound signaling molecules. It often acts as a monomer but may participate in larger complexes during functional modulation. SULF1 regulates interactions with key growth factors such as fibroblast growth factors (FGFs) and vascular endothelial growth factors (VEGFs) impacting cell communication and migration.
Pathways
SULF1 is an important regulator in signaling pathways including the VEGF signaling pathway and the FGF signaling pathway. These pathways are essential for processes like angiogenesis and embryonic development. SULF1 activity influences these pathways by modifying heparan sulfate chains affecting the binding of FGFs and VEGFs to their respective receptors alongside related proteins like SULF2 which shares similar functions in these pathways.
