    The genus Streptococcus is comprised of a wide variety of both pathogenic and commensal gram positive bacteria which are found to inhabit a wide range of hosts, including humans, horses, pigs and cows. Within the host, streptococci are often found to colonize the mucosal surfaces of the mouth, nose and pharynx. Streptococci can be divided into many groups on the basis of antigenic differences in group-specific polysaccharides located in the bacterial cell wall. More than 20 serologic groups have been identified and designated by letters, eg, A, B, C. Group A streptococci, cause a wide-range of disease in humans, from mild sore throats to life-threatening invasive disease such as necrotising fasciitis. Streptococcus pyogenes (a group A Streptococcus) is one of the most important pathogens encountered in clinical practice. Group A streptococci (GAS) are gram-positive, nonmotile, non-spore-forming organisms that appear as pairs or short-to-moderate-sized chains. Group A organisms can be identified by enzyme immunoassays.

Following up my message from yesterday, our source for this antibody confirmed that the carbohydrate that is referred to in the specificity field of the datasheet is "type specific" insofar as it is unique to group A. However, the carbohydrate/polysaccharide, a component of the cell wall, is not further characterized. I hope this helps. If you have any other questions, please contact us.

I have asked the laboratory to clarify, if possible, but I think the specificity text on the datasheet, "Type specific carbohydrate of group A Streptococcus", simply means that the antibody is reactive with an uncharacterized carbohydrate on Streptococcus group A, but is not reactive with carbohydrates isolated from other bacteria. I expect the lab will confirm that this is the case, that the carbohydrate is not further characterized or identified, but I will forward their reply when I receive it.

The datasheet says this antibody recognizes "Type specific carbohydrate of group A Streptococcus". How was this determined? Is it known if the antibody recognizes a specific carbohydrate, or just that it recognizes carbohydrates unique to group A?

The reply I received from the laboratory that produces the anti-Streptococcus group A antibody ab21034 replied to your question as follows: "Group A Streptococcus contains both group as well as type specific carbohydrate antigens in the cell wall. Our antibodies are directed to the group A carbohydrate antigen and thus will not miss any group A isolates. To do this, we used a strain that does not contain any type specific antigen and purified the group antigen from this strain. The resulting antibod(ies), once produced, are then tested to confirm that they only react with group A strep and not with any of the other strep groups." I may be able to obtain the details of the isolation of the antigen used for immunization, for instance the strain it was isolated from, but immunogen details are often considered proprietary information. Has this reply answered your question? If not, can you tell me what it is you are trying to identify? Our source may be able to tell us if the antibody is capable of identifying it.
