Hi, As you suggested, I have tried 2 more antigen retrieval methods including Tis/EDTA and proteinase k treatment. Unfortunately, non of them worked! I have no clue what else i should try!!

1) Both antibodies detect a phospho-site. Under the experimental conditions you are using is the threonine 183 phosphorylated? yes! actually based on our data on western blot for one of the ab, we expected to get the signal for phospho T183.
2) ab79199 has not been tried on mouse samples, thus we do not know if and how well the antibody works on this species. thats right but i already tried other ab which should work for mouse.
3) Have you tried other antigen retrieval methods, e.g. Tris/EDTA or enzymatic? No!
4) Do the detection systems work well with other primary antibodies? Yes, definitely!
5) Have you tried a positive control, such as human brain sections or human liver carcinoma sections?No, but, based on our data from western blot, MST1 is expressed in mouse islet of pancreas and should phosphorylated.

Since ab79199 has not been tried on mouse samples, we do not guarantee it for this species. The guarantee applies for tested species and applications only.

I would like to suggest trying other antigen retrieval methods, e.g. Tris/EDTA or enzymatic as this can help with obtaining a signal. This is the only step I can think of that might prevent you from obtaining a signal.

Inquiry: Dear Technical team, I would like to discuss about my recent problems regarding the immunohystochemistry of two abcam products: 1) Anti-MST1/MST2 (phospho T183) antibody (ab79199) 2) Anti-MST1 (phospho T183) antibody (ab63474) I have done the IHC staining of paraffin-embedded mouse pancreatic section for these proteins based on the protocol which is provided by abcam and got no signal at al! I have tried already fluorescence and bright filed staining.
Here is my experimental conditions:
1) Blocking conditions: 10% normal serum with 1% BSA in TBS for 1 hours at RT
2) Detection method: a) fluorescence b) bright-field staining based on the HRP system
3) Primary antibody conditions: dilution (1.50 or 1:100) for overnight at 4 degree
4) Sample fixation: PFA (4%) fro 6 hour
5) antigen retrieval: heat-induced epitope retrieval (Sodium Citrate Buffer)
6) permeabilization: Triton-X-100 solution (0.4%) for 30 min in ice
7) Secondary antibody conditions: a) fluorescence (Cy3 Donkey anti-rabbit from another company for 1 hour at RT) b) bright-field staining based on the HRP system
(1-other company, 1:100 Biotin anti-Rabbit ab for 1 hour at RT
2-HRP 30 min
3- HRP substrate 10 min RT)
8) Wash steps: all TBS 3 times for 2 min
at least I would like to ask for the expert comments and any help to my work.
