Immunocytochemistry/ Immunofluorescence - Anti-Staphylococcus aureus antibody (AB20920)

ab20920 staining Staphylococcus aureus in human keratinocytes by Immunocytochemistry/ Immunofluorescence.

Cells were fixed in methanol, permeabilized using 0.1% saponin/ PBS, blocked with 4% BSA for 30 minutes at 25°C and then incubated with ab20920 at 10μg/ml for 14 hours at 4°C. The secondary used was an Alexa-Fluor 488 conjugated goat anti-rabbit polyclonal used at a 1/100 dilution.

ab20920 staining an abscess in rat calf tissue prepared as a 20 μm cryoslice. Tissue was fixed in paraformaldehyde, permeabilised with 0,5% Trition X100 in PBS, and blocked with 10% serum plus 1% BSA for 1 hour at room temperature. The primary antibody was diluted 1/500 with PBS plus 3-5% Serum, 0,5% Trition X100 and incubated for 24 hours at 20°C. An Alexa Fluor® 568 goat anti-rabbit was used as the secondary

Staphylococcus aureus also known as "aureus" or "staph" is a gram-positive bacterium with a molecular mass ranging around 2.8 million daltons. It is found mainly in the nasal passages and on the skin of humans. This bacterium has a thick peptidoglycan layer making it resistant to certain environmental pressures. The structure of its proteins like Protein A plays an important role in immune evasion. Staphylococcus aureus also expresses a variety of toxins and enzymes that aid in its survival and pathogenicity.
Biological function summary

Staphylococcus aureus acts as an opportunistic pathogen that can cause infections when the host's immune system is weakened. It is not part of a specific protein complex but utilizes a range of virulence factors to establish infections. The bacterium produces biofilms which provide a protective environment against host defenses and antimicrobial agents. Its ability to quickly adapt to different environments makes it a major cause of both nosocomial and community-acquired infections.

Pathways

The metabolic activities of Staphylococcus aureus are involved in two significant pathways: the peptidoglycan biosynthesis and the toxin production pathway. Peptidoglycan biosynthesis is essential for cell wall integrity while toxin production pathways lead to the synthesis of critical virulence factors like alpha-hemolysin. Key proteins involved in these pathways include PBP2a which is significant for methicillin resistance and hemolysins that damage host tissues.

Staphylococcus aureus is connected to skin infections and pneumonia.
