補足情報

SSBP1 also known as Single-Stranded DNA Binding Protein 1 is a mitochondrial protein with an approximate mass of 14 kDa. SSBP1 binds to single-stranded DNA and stabilizes it during DNA replication and repair processes. It functions by preventing the DNA from re-annealing or forming secondary structures which helps in maintaining genomic stability. This protein is mostly expressed in tissues with high energy demands such as muscle and liver reflecting its significant role in mitochondrial activities.
Biological function summary

Single-Stranded DNA Binding Protein 1 participates in essential mitochondrial DNA maintenance processes and interacts with other proteins to form functional complexes. It associates with mitochondrial DNA polymerase gamma and other replication machinery components to ensure accurate DNA replication. These interactions are significant for proper mitochondrial function since they prevent mutations that could compromise cellular energy production.

Pathways

SSBP1 engages actively in the mitochondrial DNA replication and repair pathway. It works closely with proteins like TWINKLE helicase and DNA polymerase gamma complex facilitating efficient replication and repair of mitochondrial DNA. These pathways are pivotal for mitochondrial biogenesis and function impacting cellular energy metabolism and overall mitochondrial health.

Alterations in SSBP1 have links to mitochondrial dysfunctions including disorders like mitochondrial myopathy and progressive external ophthalmoplegia. Dysfunctional SSBP1 may lead to impaired DNA replication or repair in mitochondria contributing to these conditions. Additionally it relates to TWINKLE helicase mutations which also disrupt mitochondrial DNA maintenance further emphasizing the importance of SSBP1 in maintaining cellular energy balance.

Binds preferentially and cooperatively to pyrimidine rich single-stranded DNA (ss-DNA) (PubMed : 21953457, PubMed : 23290262, PubMed : 31550240). In vitro, required to maintain the copy number of mitochondrial DNA (mtDNA) and plays a crucial role during mtDNA replication by stimulating the activity of the replisome components POLG and TWNK at the replication fork (PubMed : 12975372, PubMed : 15167897, PubMed : 21953457, PubMed : 26446790, PubMed : 31550240). Promotes the activity of the gamma complex polymerase POLG, largely by organizing the template DNA and eliminating secondary structures to favor ss-DNA conformations that facilitate POLG activity (PubMed : 21953457, PubMed : 26446790, PubMed : 31550240). In addition it is able to promote the 5'-3' unwinding activity of the mtDNA helicase TWNK (PubMed : 12975372). May also function in mtDNA repair (PubMed : 23290262).
