    Receptor-regulated SMAD (R-SMAD) that is an intracellular signal transducer and transcriptional modulator activated by TGF-beta (transforming growth factor) and activin type 1 receptor kinases. Binds the TRE element in the promoter region of many genes that are regulated by TGF-beta and, on formation of the SMAD2/SMAD4 complex, activates transcription. May act as a tumor suppressor in colorectal carcinoma.

    Phosphorylated on one or several of Thr-220, Ser-245, Ser-250, and Ser-255. In response to TGF-beta, phosphorylated on Ser-465/467 by TGF-beta and activin type 1 receptor kinases. Able to interact with SMURF2 when phosphorylated on Ser-465/467, recruiting other proteins, such as SNON, for degradation. In response to decorin, the naturally occurring inhibitor of TGF-beta signaling, phosphorylated on Ser-240 by CaMK2. Phosphorylated by MAPK3 upon EGF stimulation; which increases transcriptional activity and stability, and is blocked by calmodulin.
    In response to TGF-beta, ubiquitinated by NEDD4L; which promotes its degradation.
    Acetylated on Lys-19 by coactivators in response to TGF-beta signaling, which increases transcriptional activity. Isoform short: Acetylation increases DNA binding activity in vitro and enhances its association with target promoters in vivo. Acetylation in the nucleus by EP300 is enhanced by TGF-beta.

    Question

    I did WB for phospho Smad2 using your company's antibody. I got the strong bands after 3 minutes development with ECL, but MW is higher than the data sheet. (58kDa in data sheet, but I got around the 90-100kDa). I did the long exposure for 2hours, but the bands around 60kDa is very faint. In the data sheet, there is no extra bands around 90-100kDa. Do you think the 90-100kDa bands are extra bands and 69kDa faint bands are true bands? Or 90-100kDa bands are true bands?? Or are there any troubles in antibody?? The picture is attached to this e-mail (2hr exposure). The WB condition is below. •Amount of protein loaded 20ug NRK-49F cells (rat kidney fibroblast cell line), total cell lysate •Blocking conditions 5% skim milk in TBS-T •Electrophoresis 100V 120min transfer conditions 400mA 60min •Primary antibody conditions ab53100 400:1, 4C, Over night (16hr approx) •Secondary antibody conditions PerkinElmer, ECL solution 1000:1, room temp 2hr •Wash steps 10min x 6times with TBS-T

    Thank you for getting back to me. I am interested in measuring Smad2
    that has been phosphorylated by the TGF-beta type I receptor. I
    believe that this phosphorylates serine 465 and serine 467, so I am
    guessing that product#ab53100 may work for either the capture or
    detection.

    Answer

    Thank you very much for your reply. My colleague is out of the office this week, but I'll be happy to help you with this enquiry.

    You could use ab53100 in a sandwich ELISA to study the SMAD2 protein phosphorylated at residue S467. I'd suggest using this as the detection antibody and a total SMAD2 antibody as the capture antibody. Ab47083 is a total SMAD2 antibody that has been tested for reactivity with bovine samples, so this may be a good choice-

    Since this is also a rabbit polyclonal antibody, you would need to directly conjugate the detection antibody to HRP or AP because an anti-rabbit secondary antibody will bind to both of the primary antibodies. We have conjugation kits which can be used for this purpose-

    Another option would be to use a mouse monoclonal total SMAD2 antibody that hasn't been tested with bovine samples, since the protein is believed to be fairly conserved and it might cross-react with your samples. In this case, you could use a pre-adsorbed anti-rabbit secondary antibody like ab7090 that won't cross-react with the mouse capture IgG. We have ab119907 which could be used for this-

    Since we haven't tested ab119907 or ab53100 with bovine samples, we can't guarantee that they will cross-react and work in your assay.
