SET3 also known as SET domain containing 3 is a protein with a molecular mass of approximately 98 kDa. It functions as a histone methyltransferase an enzyme that adds methyl groups to histone proteins. This action influences chromatin structure and gene expression. SET3 is expressed in various tissues including heart and skeletal muscle with notable levels in hematopoietic cells. It plays an essential role in regulating the transcriptional programs inside these cells.
Biological function summary

In the cell SET3 participates as part of a multi-protein complex known as the SET3 complex. This complex is involved in deacetylating and methylating histones which affects gene transcription levels. The modification of histones managed by the SET3 complex helps in maintaining stable gene expression patterns necessary for cell differentiation and development processes. It ensures correct chromatin organization and therefore proper transcription regulation.

Pathways

Histone modification processes involve SET3 contributing significantly to gene silencing pathways and chromatin remodeling. In gene silencing pathways SET3 works closely with histone deacetylases to remove acetyl groups therefore repressing gene expression. Chromatin remodeling involves consort with other proteins like SIN3A to control access of transcription factors to DNA. These pathways are essential for proper embryonic development and cell identity maintenance.

Disruption of SET3 expression can link to various hematological malignancies like leukemia where chromatin misregulation plays a role. Its aberrant function or expression might also connect to developmental disorders due to failure in proper gene regulation. In leukemia SET3 correlates with proteins like RUNX1 and CEBPA which are critical for hematopoietic differentiation potentially leading to hematopoietic cell transformation when improperly regulated.
