Western blot - Anti-SEL1L antibody (AB78298)
This blot was produced using a 4-12% Bis-tris gel under the MOPS buffer system. The gel was run at 200V for 50 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 2% Bovine Serum Albumin before being incubated with ab78298 overnight at 4°C. Antibody binding was detected using an anti-rabbit antibody conjugated to HRP, and visualised using ECL development solution ab133406.
SEL1L has a predicted molecular weight of 89 kDa; however it has a number of potential glycosylation sites which may affect the migration of the protein (SwissProt data). This migration has been observed in the literature (PMID : 18502753).

Biological function summary
The SEL1L protein is critically involved in the ER-associated degradation (ERAD) pathway where it works as part of a complex with other proteins like HRD1. This role allows SEL1L to contribute to the removal of misfolded or damaged proteins from the ER preventing cellular toxicity and dysfunction. In the context of protein quality control SEL1L ensures that only correctly folded proteins proceed to their functional destinations thereby maintaining cellular environment stability and function.
Pathways
SEL1L integrates into the unfolded protein response (UPR) and ERAD processes. The function of SEL1L within these pathways links it to key proteins such as HRD1 and BiP facilitating a critical response mechanism when unprocessed or misfolded proteins accumulate within the ER. By participating in these pathways SEL1L helps in regulating cellular stress responses and preserving protein homeostasis contributing to the proper function of the secretory pathway and cellular stress adaptation.
